Malignant tumor caused serious damage to the human health,melanoma and lung cancer were two of the most deadly tumors so far.Malignant tumor was very difficult to eradicate by using traditional treatments.With the continuous development of tumor immunotherapy,the method improving tumor microenvironment got more attention from researchers.To explore the antitumor effects and the potential mechanisms,we studied the therapeutic effect of Tanshinone?,Aspirin combined with Metformin on malignant melanoma and Lewis lung cancer in mice by using different treatments.B16F10 cell line and Lewis lung cancer cell line were used,subcutaneous transplantation tumor model was established to discuss the antitumor effect.In the course of the experiment,the body weights and tumor diameter were measured.The lung metastasis was observed,RT-qPCR was used to detect the mRNA of metastasis associated proteins in the lung.Tumor infiltrating lymphocytes were detected by flow cytometry.The change of inflammatory factors such as interleukin-6(IL-6),interleukin-1?(IL-1?),tumor necrosis factor?(TNF-?)were detected by ELISA.Results showed that the combination of three drugs could inhibit the growth of melanoma(P<0.01),decrease the wet weight of tumor(P<0.01),suppress tumor lung metastasis.It's antitumor effect might be caused by increasing the tumor infiltrating CD8~+T lymphocyte and the changes of the inflammatory factors in tumor microenvironment.Besides,it didn't have any effect on Lewis lung cancer either in terms of inhibiting tumor growth or lung metastasis.The optimum concentration of the drug was screened by the influence of different drug gradient on the proliferation of B16F10 cell and Lewis lung cancer cell.The proliferation of two cell lines were observed under the treatment of different drugs.The results showed that the B16F10 cells were more sensitive to these three drugs than the Lewis lung cancer cell.In addition,the IL-6 expression of the two cell lines was tested after 24h and the results showed that the IL-6 expression of B16F10 cell was 1.59 times than that of Lewis lung cancer cell.Therefore,it is possible to infer that the two different outcomes of animal levels may be related to the expression of IL-6.
