Objective To explore the association between TRPV5 polymorphism and high blood lead levels.Methods1.The concentrations of lead in the workplace air were measured.2.All the subjects underwent uniform questionnaire survey on daily habits and took the measurement of blood lead levels and Zinc Protoporphyrin(ZPP).3.To assess the factors which influence blood lead levels of workers who occupationally exposed to lead,and the association between ZPP and blood lead.Using benchmark dose calculation software(BMDS)to calculate blood lead benchmark dose and dose lower limit of the 95%confidence interval(BMDL)by applying blood lead as exposure marker and ZPP as effective marker.4.We selected 8 SNPs based on NCBI and articles.Furthermore,we determined the genotypes of eight single-nucleotide polymorphisms(SNPs)in 226 lead-sensitive and 226 lead-resistant Chinese occupationally exposed to lead by Taqman assay.Results1.Of the five companies,the percent of lead fume in the workplace maintained below the limited value was 75%,while percent of the lead dust was 69.2%.2.Statistical differences were observed among the three groups on blood lead,age,gender,education levels,type of work,smoking and drinking status in univariate analysis.Age,gender,education levels,and external exposure degree of lead,and smoking were the influence factors of high blood lead in Logistic regression analysis.There was significant(P<0.001)positive linear correlation between ZPP and blood lead in certain extent.Blood lead benchmark dose and BMDL were 304.29p,g/L and 285.34?g/L by using blood lead as exposure markers and ZPP as effective markers(P=0.089).3.One SNP in TRPV5(rs4252416)was significantly associated with lead-poisoning(P= 0.034.OR = 0.33,95%CI =0,12-0.92 in the GT genotype co mpared to the TT genotype).There were no significant associations between th e other seven SNPs and the blood lead levels.Conclusion1.For the workers in the manufactures,age,gender,and external exposure degree of lead,and smoking were the influence factors of high blood lead.Under the condition of high blood lead level,blood lead and ZPP have high correlation and ZPP is a sensitive indicator of lead exposure.In actual work,when using ZPP as indicator of low blood lead level(<100?g/L),other indicators should be combined with ZPP.2.Therefore,polymorphism rs4252416 could be used to distinguish lead-resistant and lead-susceptible populations,and to prevent the lead-related workers.
