Purpose:In this paper,the application of modern polysaccharide extraction technology for the extraction and separation of Eupolyphaga sinensis polysaccharide.Then the extraction effect was analyzed,and the content of polysaccharide was determined,and the effect of inhibiting the tumor in mice was discussed.This study can be helpful to the comprehensive utilization of insect resources,and can provide reference for the study of insect functional food and drugs.Methods: after the Eupolyphaga sinensis raw material pretreatment,hot water extraction method for extracting polysaccharide from the extraction,alkali extraction and enzymatic hydrolysis of dilute.Then,the influence of various factors on the extraction of polysaccharides was compared,and the optimum extraction process of polysaccharides was determined on the basis of orthogonal experiments.The phenol sulfuric acid spectrophotometric method of Eupolyphaga sinensis polysaccharide content were determined and analyzed the content of polysaccharide.The effect of extracted polysaccharides on inhibiting immune function and S-180 tumor growth in mice was studied,and the mechanism of action was discussed.Results: the effect of alkali extraction is the best,the content of polysaccharide is the highest,and the content of polysaccharide is 24.31%.The optimum extraction conditions of Eupolyphaga sinensis polysaccharide is: alkali concentration corresponding to 0.03 mol/L,the extraction temperature corresponding to 60 degree,extraction time was 1 h.Eupolyphaga sinensis polysaccharide can increase the weight of immune organ,the influence mechanism is complicated.After feeding the polysaccharide in mice,the thymus and spleen indices of mice were improved,and the clearance index and corrected phagocytic index of mice were significantly improved compared with those before the experiment.Can be judged accordingly Eupolyphaga sinensis polysaccharide can play a non-specific immune function in immune regulation,in addition to its side effect is significant.According to the above comparison,the polysaccharides effectively inhibited the growth of S-180 sarcoma in mice in each dose conditions,and in the 200 ~ 600mg/ kg/d range of inhibition rate and dose are positively correlated,reached a high level,under the condition of 600 mg/kg/d can get the best inhibition level,and the corresponding suppression the tumor rate reached 50.48%.Conclusion: the extract of Eupolyphaga sinensis polysaccharide extraction method effect has obvious difference.The best dilute alkali extraction effect,the extraction rate and extraction rate was significantly higher than that of hot dipping extraction and hydrolysis extraction method.This method is easy to operate and low in cost.It can meet the requirement of polysaccharide extraction well and has certain popularization and application value..
