Objective:Exosome are nanoside extracelluar vesicles released to extracellular milieu through fusion of multivesicular bodies with the plasma membrane,whose size is range from 30-150 nm,these exosomes contain micro RNAs and might be transferring information between cells.The aim of this study was to analysis the effect of proliferation and apoptosis of CD LNCAP cells which is treated by exosomes of CAF cells and to analysis exosomal micro RNAs of proliferation and apoptosis in CAF cells.Methods:Extract the exosomes from CAF cells by ultracentrifugation method.Using transmission electron microscopy,nanometer particle tracking analysis and Western Blot to identify the exosomes.Using MTT,cell cycle analysis,cell count and cell apoptosis to identify the effect of proliferation and apoptosis of CD LNCAP cells which is treated by exosomes of CAF cells.Using trizol method to extract the RNA of CAF cells and exosomes and RNA of NF cells and exosomes and identify micro RNA by q PCR and PCR.Results:Electron microscopy of exosomes reveals cup-shaped vesicles.Exosomes of CAF cells line contains CD63 protein,distribution peak is about 100 nm,ranging from 30-150 nm.Using MTT,cell cycle analysis,cell count and cell apoptosis can identify that exosomes of CAF cells promote proliferation and inhibit apoptosis of CD LNCAP cells.Using q PCR and PCR can identify that exosomes of CAF cells contain mi R-21 which promotes the growth of prostate cancer.Conclusions:CAF cells can secrete exosomes that rang from 30 ~ 150 nm in diameter,which express CD63 protein.Exosomes of CAF cells can promote proliferation and inhibit apoptosis of CD LNCAP cells.Exosomes of CAF cells contain mi R-21 which promotes the growth of prostate cancer and maybe it induce the effect of proliferation and apoptosis.
