| ObjectiveOvarian cancer is the deadliest of all gynecological malignancies,5-year survival rate of less than 30% and poor prognosis.Ovarian serous cancer is the most common malignant tumor of ovarian epithelial carcinoma,the mainly reason are the high aggressive,recurrence,drug resistance.According to our previous work,we found that DSG2 may play a role in malignant transformation of cancer stem cells.At the same time by reading the literatures found that DSG2 may also be involved in the development of skin cancer,breast cancer,gastric cancer and other malignant tumor occurrence;However,DSG2 in ovarian cancer,especially the expression of ovarian epithelial carcinoma.There is no clear data.Therefore,this study will be carried out the expression of DSG2 in ovarian serous tumors:1.To study the expression of DSG2 protein in ovarian serous cancer.2.To explore the expression of DSG2 protein in HGSC.Methods1.Compare the expression of DSG2 gene m RNA in ovarian serous carcinoma: According to the two-system theory,selection of ovarian serous tumor specimens,namely SCA,SBT,LGSC and HGSC.Detecting the expression and significance of DSG2 gene m RNA in ovarian serous cancer of four groups by real-time quantitative PCR(qRT-PCR).2.To clarify Association of DSG2 expression in high-grade serous ovarian cancer and the clinical and pathological characteristics:Using immunohistochemistry(S-P)method to detect the expression of DSG2 protein in ovarian serous carcinoma.The positive expression of DSG2 was located on the cell membrane,which showed the yellow or brown granules.Scoring criteria:(1)calculate the percentage of positive cells in total cells,<25%=1 score,25%-50%=2 score,50%-75%=3 score,>75%=4 score;(2)staining intensity score: weak =1 score,strong =2 score,stronger =3 score.Add(1)to(2),that is,DSG2 score.3.To investigate the expression and significance of DSG2 protein in high-grade serous ovarian cancer:Using immunohistochemistry(S-P)method to detect the expression of DSG2 protein in HGSC and the correlation of clinical pathological parameters.Results1.The expression of DSG2 in ovarian serous cancer(1)The expression of DSG2 gene m RNA in ovarian serous cancerThe expression trend of DSG2 gene m RNA in 33 cases of ovarian serous carcinoma : SCA,SBT,LGSC,HGSC were increasingly up-regulated.(2)The expression of DSG2 protein in ovarian serous cancerThe expression of DSG2 protein in 105 cases of ovarian serous cancer.The results showed that DSG2 protein in HGSC was lower than that of SBT expression(P<0.05);the expression of DSG2 protein in HGSC was lower than that of LGSC(P<0.05).2.The expression of DSG2 protein in HGSC(1)The expression of DSG2 protein in HGSC was detected by IHCContinuous and weekly positive expression of DSG2 protein in the cell membrane of the normal fallopian tube epithelium.The results showed that the expression of DSG2 protein in 34 cases of HGSC were absent in part of epithelial,and the expression of cell membrane was not complete,there were some cases of vague expression in cytoplasm.While the relationship between cell morphology and expression of DSG2 protein showed HGSC cell morphology of Papillary was higher than the level of Solid sheet cell morphology(P<0.05).In addition,observed in 19 patients with HGSC,the expression of DSG2 protein in the area around the Papillary while in central region micropapillary.Found from 11 cases of HGSC,the expression regional intensity of DSG2 protein in SBT,LGSC and HGSC became strong-stronger-weak.(2)The relationship between the expression of DSG2 protein and prognosis(2)The relationship between the expression of DSG2 protein and prognosis in patients with HGSCThe higher expression of DSG2 protein in HGSC,the better the overall survival(P<0.05)and progression free survival(P<0.05).(3)Association of DSG2 expression levels in HGSC and mitotic figuresHigh expression of DSG2 mitotic figures in HGSC was less than low expression group(P<0.05).(4)Association of DSG2 protein and morphology in HGSCThe expression of DSG2 protein in Papillary in HGSC was higher than that in Solid sheet.Conclusions1.DSG2 gene may be involved in malignant transformation of ovarian carcinoma.2.Low expression of DSG2 protein has a certain reference value for the prognosis in HGSC. |
