Objective:1.By immunohistochemical detection the expression of FOXO1 and its downstream molecules KLF2 and S1P1 in thymoma associated with myasthenia gravis(MG),thymic hyperplasia,simple thymoma and normal thymus four differents tissues,and know the significance clinical of their expression in different tissues.2.FOXO1 and its downstream molecules KLF2 and S1P1 were detected in patients with thymoma associated with myasthenia gravis,and analyze the relationship with disease duration,age,sex,histological type,clinical stage and Ach R-Ab.3.To investigate the correlation with foxo1-klf2-s1p1 signal axis in tissue specimen and to analyze the relationship between signal axis and disease.2.Methods:1.Immunohistochemical method(SP method)was used to detect the expression of FOXO1 and its downstream signaling molecules KLF2 and S1P1 in different sections of thymus.There were 22 cases of thymoma associated with MG,thymic hyperplasia in 5 cases,simple thymoma in 7 cases and normal thymus tissue in 4 cases.2 the expression levels of FOXO1,KLF2 and S1P1 in different thymic tissues were detected by real-time fluorescence quantitative PCR(m RNA).3 Statistical software SPSS22.0 was used to analyze the correlation between the expression of FOXO1,KLF2 and S1P1 in different thymus tissues and the related factors.2.Results:1.FOXO1 in MG with thymoma and thymic hyperplasia and simple thymoma patients were positive,the positive rate was 77.3%(17/22),80%(4/5),42.8%(3/7),;in the normal thymus was 25%(1/4).The expression of FOXO1 in thymoma associated with MG was significantly higher than that in pure thymoma(P < 0.05),but there was no significant difference between thymus hyperplasia and MG(P >0.05).In the cases of thymoma with MG,FOXO1 is mainly expressed in the medulla and the surrounding islands;the nucleus and cytoplasm in thymic hyperplasia is mainly expressed in the cortex and medulla area,was mainly in nuclear;scattered expression in normal thymus tissue.2.KLF2 in MG with thymoma and thymic hyperplasia and simple thymoma patients were positive,the positive rate was 86.3%(19/22),100%(5/5),85.7%(6/7),;in the normal thymus tissues the expression rate was 25%(1/4).In thymoma with MG,the expression of KLF2 was significantly higher than that of pure thymoma(P < 0.05)and normal thymus tissue(P < 0.05),but there was no significant difference between thymus hyperplasia and MG(P > 0.05).With KLF2 MG in thymoma was mainly expressed in the medulla and the surrounding Island,the high expression in the cytoplasm;while in thymic hyperplasia mainly expressed in cortex,medullary junction and medulla;normal thymus tissue and scattered expression in.3.S1P1 in MG with thymoma and thymic hyperplasia and simple thymoma patients were positive,the positive rate was 81.8%(18/22),80%(4/5),42.8%(3/7);the normal thymus is low expression in thymoma with MG;the expression of S1P1 in high and normal thymus tissue was significant difference(P < 0.05).In the tissues of thymoma,S1P1 was mainly distributed in the medulla islands and perivascular spaces,while in the thymus hyperplasia specimens were mainly distributed in the medulla.4 In the thymoma with MG tissue,the relative expression of the three proteins was4.32±0.86 ? 3.58±0.86 4.06±0.67 respectively.In simple thymoma,the relative expression of three proteins was 45.99±1.50,5.72±0.89,5.85±1.25.In thymus hyperplasia,the relative expression of three proteins was4.38±0.25,4.18±0.55 ?4.23±0.476.The relative expression of FOXO1,KLF2,S1P1 in thymoma combined with MG was higher than that of thymoma(P < 0.05)5.Correlation analysis of FOXO1,KLF2,S1P1,FOXO1 and KLF2 in thymoma associated with the expression of MG(=0.497,P=0.019<0.05),see figure 16.FOXO1 and S1P1 thymoma associated with the expression of MG(=0.481,P=0.043<0.05),see figure 16.1.There was no correlation between the expression of KLF2 and S1P1 thymoma with MG(=4.286,P=0.143>0.05),as shown in figure 16.2;6.There was no significant correlation between the expression of FOXO1,S1P1 and KLF2,such as course of disease,age,sex,histological type and clinical stage Ach R-Ab.Conclusion:1?The expression levels of FOXO1,KLF2 and S1P1 were increased in patients with thymoma associated with MG and thymus hyperplasia,which indicated that the pathogenesis of MG may be related to the pathogenesis of MG and the abnormal foxo1-klf2-s1p1 signal axis.2?The expression of FOXO1,KLF2,and S1P1 in thymoma and thymus hyperplasia were different,shows that their pathogenic mechanism is different.3 ? In thymoma with MG,FOXO1 and the downstream molecules KLF2 ? S1P1 m RNA,the relative expression was higher than simple thymoma,indicating that the foxo1-klf2-s1p1 axis may affect the output of abnormal thymus cells,and control the number of peripheral T cells.
