| Objective:To investigate the expression of LMP1 protein and Cripto-1 protein in 42 nasopharyngeal carcinoma tissues and 22 nasopharyngeal tissues,explore the role of LMP1 protein and Cripto-1 protein in the development of nasopharyngeal carcinoma then investigate the correlation between them;analysis the LMP1 and Cripto-1 in nasopharyngeal carcinoma cells and to investigate the effect of LMP1 on the propagation of nasopharyngeal carcinoma cells,particularly in the potential regulatory mechanisms。 Method: 1、 In this study,we use the immunohistochemistry to evaluate the expression of LMP1 andCripto-1 in the 42 patients of nasopharyngeal carcinoma and 22 patients with nasopharyngitiswere used as control;the expression of LMP1 protein and Cripto-1 protein in different nasalpharyngeal disease,different pathological types 、 sex 、 age,whether lymph node hasmetastasised and TNM staging in different nasopharyngeal carcinoma,explore the correlationbetween them。 2、The proliferation of CNE1 cells and CNE1-LMP1 cells were detected by CCK8。 3、Western Blot 、RT-PCR methods were used to tested the expression of LMP1 and Cripto-1 in different nasopharyngeal carcinoma cell lines。 Results: 1、 Through the detection of nasopharyngeal mucosa specimens in the localization andexpression of LMP1 protein levels found that LMP1 positive expression rate innasopharyngitis tissue was only 22.73%,uniform distribution,then in nasopharyngealcarcinoma tissue positive expression rate was 73.81%,the expression of different levels incancer cell membrane and(or)cytoplasm。Furthermore,divided nasopharyngeal carcinomatissues into cases with or without lymph node metastasis,LMP1 expression in the two groupshave statistically significant differences(P < 0.01),the former positive rate was 86.67%,thelatter was only 41.67%。In addition,the expression of LMP1 protein in different types、gender 、age of of nasopharyngeal carcinoma patients had no significant difference(P>0.05);correlated with TNM stage of tumor(P <0.05)。 2、 Cripto-1 protein was also mainly expressed in the membranes and(or)cytoplasm ofnasopharyngeal carcinoma cells,positive expression was mainly concentrated in thedistribution of nasopharyngeal carcinoma cells,in nasopharyngitis tissues the positiveexpression of Cripto-1 protein was lower。The positive rate of Cripto-1 in nasopharyngealcarcinoma patients and nasopharynx pharyngitis patients were 80.95% and 36.36%repectively;Furthermore,divided nasopharyngeal carcinoma tissues into cases with orwithout lymph node metastasis,Cripto-1 expression in the two groups have statisticallysignificant differences(P < 0.01),the former positive rate was 90.33%,the latter is only 50%。In addition,the expression of Cripto-1protein in different types 、 gender 、 age of ofnasopharyngeal carcinoma patients had no significant difference(P>0.05);correlated withTNM stage of tumor(P <0.05)3、CNE1-LMP1 cell line had stronger proliferation ability than CNE1 cell line(P <0.01)。 4、CNE1-LMP1 cell line stably express LMP1 gene,Cripto-1 expression in protein and mRNA level were dramatically higher than that of CNE1 cells,the difference was statistically significant(P < 0.05)。 Conclusion: 1、 LMP1 and Cripto-1 expression in nasopharyngeal carcinoma patients were significantlyhigher than those in nasopharyngitis patients,It is suggested that LMP1 and Cripto-1 wereimportant in the development,progression and metastasis of nasopharyngeal carcinoma。 2、 The expression of LMP1 and Cripto-1 in nasopharyngeal carcinoma is positive related。 3、 LMP1 can adjustable the expression of Cripto-1 in different nasopharyngeal carcinoma cell;the expression of LMP1 is closely relevant to the proliferation of nasopharyngeal carcinomacells,which indicates expression of LMP1 was closely related to invasion and metastasis ofnasopharyngeal carcinoma。It could be used as a molecular signs for screening,diagnosisand intervention of nasopharyngeal carcinoma 。... |
