Objective: With relieving mechenical withdrawal threshold,improving the motor nerve conduction velocity,sciatic nerve morphology,neurobiochemical markers and cells apoptosis in STZ-induced DPN rats.In order to have a more comprehensive evaluation of koumine on the treatment of DPN,for its development and application to provide experimental basis and theoretical basis.Methods:(1)SD rats were randomly divided into control group,model group and koumine(1.4 mg/kg and 0.28 mg/kg)group in STZ-induced DPN rats.The day of STZ administration was regarded as day 0.Mechanical withdrawal threshold was measured before STZ administration,and post-administration days 34 and 37,39,41.To observe the effect of KM on mechenical withdrawal threshold in DPN rats.(2)The motor nerve conduction velocity was measured on post-administration day 42.The effect of KM on the motor nerve conduction velocity in DPN rats was observed by neurophysiological method.(3)After the motor nerve conduction velocity was measured,the sciatic nerve was dissected on day 43 and the morphology assessed with toluidoine blue staining and transmission electron microscopy.(4)After the motor nerve conduction velocity was measured,the sciatic nerve was dissected on day 43 and the levels of MBP and NFP assessed with immunohistochemistry.(5)After the motor nerve conduction velocity was measured,the sciatic nerve was dissected on day 43 and the cells apoptosis assessed with TUNEL assay.Results:(1)Behavioral results showed that KM could significantly improve the mechenical withdrawal threshold in DPN rats.(2)Neuroelectrophysiological results showed that KM could improve the motor nerve conduction velocity in DPN rats.(3)The results of toluidine blue staining and transmission electron microscopy indicated that sciatic nerve showed significant demyelination changes and axon atrophy in the control group.(4)The results of immunohistochemistry showed that koumine could improve the levels of MBP in sciatic nerve of DPN rats(P <0.05),and the effect on the levels of NFP in sciatic nerve of DPN rats was not clear.(5)The effects of koumine on the cells apoptosis in sciatic nerve assessed with TUNEL assay in STZ-induced DPN rats is positive.Conclusion: This is the significant effect of koumine on DPN.Koumine can improve the mechenical withdrawal threshold,motor nerve conduction velocity and morphology of sciatic nerve.What'more,koumine could improve the levels of MBP in sciatic nerve of DPN rats.The effects of koumine on the cells apoptosis is positive.
