Objective:To investigate the different expression of Sox4 during palate development and cleft palate formation of BALB/c mice,and explore the function of Sox4.Methods:The BALB/c pregnant rats were randomly divided into two groups.Embryonic mice were treated with 50mg/kg retinoic acid(RA)in the experimental group,the control group was given equal amount of corn oil.The mice were obtained on the 17 gestation day(GD17).The form of palates were Observed Under the magnifying glass.The embryonic mice in experimental group and control group were obtained on the13 gestation day(GD13),GD14,GD15.The form of palates were observed by HE stain and the expression of Sox4 were detected by immunohistochemistry.Results:The incidence of cleft palate in the experimental group was treated with 50mg/kg RA was 84.6%.Semi quantitative detection of Sox4 protein by optical density told us that the optical density of GD14 was largest in control group.We can observe that epithelium of palates began to contact and fuse with each other,which created medial epithelial seam(MES)in control group on GD14.While the palates of mice in experiment group didn't fuse all the way by HE staining.Through the comparison of optical density between experimental group and control group we found there was no difference on GD13,the expression in control group was higher than that in experiment group on GD14,which was different from the results on GD15.Conclusion:The expression of Sox4 was different between control group and experimental group on GD14-GD15.it was highest in MEE on GD14.Sox4 might play a role in palatal fusion period.
