Objective:1.To observe the effects of telmisartan on the fasting plasma glucose(FPG),fasting insulin(FINS),triglyceride(TG),homeostasis model assessment insulin resistance index(HOMA-IR)in rats induced by long high sugar and high fat diet.2.To observe the effects of telmisartan the express of protein tyrosine phosphatase-1B(PTP-1B)in islet of rats induced by long high sugar and high fat diet.To discuss the possible of telmisartan improve the insulin resistance.Methods:A total of 30 male SD rats aged from 6 to 8 weeks(180-200g)were randomly assigned into 3 groups of control group(n=10),high sugar and high fat group(n=10)and telmisartan group(n=10).The control group was given normal diet and the other groups were given high sugar and high fat diet for 24 weeks.The rats of telmisartan group were treated with telmisartan(10mg/Kg/d,dissolved in 2 ml physiological saline to lavage)for12 weeks.Index detection : After telmisartan intervention 12 weeks,The rats were anesthetized with urethane(4 ml/kg)after fasted for 8-12 hours.Taking blood from abdominal aortic blood 1 ml.Plasma was collected by centrifugation 3000 r/min,20 min.to test FPG and TG,FPG was measured by glucose oxidase method.TG was measured using by automatic biochemistry instrument.FINS was analyzed by ELISA.Insulin resistance index was calculated HOMA-IR,(HOMA-IR = FINS × FBG / 22.5).Immunohistochemistry analysis: the pancreatic tissue was obtained and was put into the 10% neutral formalin to be fix,embedded in paraffin,2 um Sections were mounted onglass slides,the pancreatic tissue were stained immunohistochemistry technique to qualitatively analyze the relative content of PTP-1B using optical microscopy combined scanscope digital pathology scanning system.Results:1.After telmisartan intervention 12 weeks,in compared with control group,the FPG level in high sugar and high fat group was higher(P<0.05).In compared with high sugar and high fat group,the FPG level in telmisartan group was lower(P<0.05).2.After telmisartan intervention 12 weeks,in compared with control group,the FINS level in high sugar and high fat group was higher(P<0.05).In compared with high sugar and high fat group,the FINS level in telmisartan group was lower(P<0.05).3.After telmisartan intervention 12 weeks,in compared with control group,the TG level in high sugar and high fat group was higher(P<0.05).In compared with high sugar and high fat group,the TG level in telmisartan group was lower(P<0.05).4.After telmisartan intervention 12 weeks,in compared with control group,the HOMA-IR level in high sugar and high fat group was higher(P<0.05).In compared with high sugar and high fat group,the HOMA-IR level in telmisartan group was lower(P<0.05).5.After telmisartan intervention 12 weeks,the pancreatic tissue were stained immunohistochemistry technique to qualitatively analyze the relative content of PTP-1B.In compared with control group,the relative expression of PTP-1B in high sugar and high fat group was increased(P<0.05).In compared with high sugar and high fat group,the relative expression of PTP-1B in telmisartan group was decreased(P<0.05).Conclusion:1.The level of FPG,TG,FINS,HOMA-IR and the relative expression of PTP-1B were increased in rats induced by long high sugar and high fat.2.After telmisartan intervention 12 weeks,the level of FPG,TG,FINS,HOMA-IRand the relative expression of PTP-IB in telmisatan group was decreased.the telmisartan improve the insulin resistance was supposed to related to the decrease of the relative expression of PTP-1B.
