Objective:Investigate the association of BMP7(morphogenetic protein 7,BMP7)gene mutation and type 2 diabetes mellitus a in Han Nationality in Taiyuan area.From the level of molecular genetics to clarify genetic mechanisms of why Han Nationality in Taiyuan are predisposed to diabetes genetic mechanisms,and provide genetic evidence for prevention.Methods:Choose on the basis of epidemiological investigation of case-control study,173(male 104,female 69)Han Chinese in Taiyuan area as the research object,according to the WHO in 1999 established diagnostic criteria for type 2 diabetes,will be the research object is divided into diabetic group(90 cases,male 56 cases,female 34 cases)and normal control group(83,men 48 and women 35).Selected two representative BMP7 gene loci in research object SNaPshot-PCR genotype identification,by hexokinase method determination of fasting blood glucose(FBG,mmol/L),chemiluminescence determination of fasting blood insulin(FlNS,mIU/L),the steady-state model assessment method(HOMA insulin resistance index(HOMA-IR=FPG×FINS/22.5),beta cell function index(HOMA-?)with HOMA-beta = 20×FINS/(FBG-3.5)evaluation,and analysis the link of BMP7 gene mutation and diabetes and insulin resistance.Results:1.The rs6025422 and rs17480735 of BMP7 gene are according with the law of equilibrium by the Hardy-Weinberg test(P > 0.05).The rs6025422 gene distribution are statistically different(P < 0.05),but the rs17480735 gene distribution are not statistically significant in the normal control group and the diabetes group by the Chi square test(P >0.05).2.Logistic regression analysis show that the GG genotype of rs602542 is a protective factor for diabetes,and the risk factors of diabetes are age and BMI.3.About the diabetes phenotype data in male comparison shows that HOMA-IR,fasting blood glucose,FINS have significant difference in the gene of rs6025422 among different genotype groups(P<0.05),but the HOMA-? has not significant difference(P>0.05).The difference in fasting blood glucose and HOMA-IR positive P value still show statistically significant(P?< 0.05).By each other comparing show that fasting blood glucose,FINS,HOMA-IR have a decreasing trend in AA,AG and GG genes.FINS and HOMA-IR have significant difference in the total population,but HOMA-? has no difference(P > 0.05).All of the data is not different in female population.Conclusion:BMP7 gene mutation may be related to type 2 diabetes and insulin resistance of Han Nationality in Taiyuan area.
