ObjectiveParthanatos is a new term of cell death similar to apoptosis,which is associated with PARP-1 and AIF.Through middle cerebral artery ischemia-reperfusion model of SD rats which was injected intraperitoneally with NBP,observing the expression levels of PARP-1and AIF and the brain morphology,to explore the neuroprotective mechanism butylphthalide in cerebral artery occlusion reperfusion injury on Sprague-Dawley rats.MethodsTirty-six male SD rats were randomly divided into the Sham-operated group,the model group(cerebral ischemic reperfusion group)and the butylphthalide treatment group(treatment group),12 rats were included in each group.The middle cerebral artery occlusion was established by the modified Longa suture method and reperfusion was initiated after 2 hours.In the treatment group,a single dose of 80mg/kg butylphthalide was injected intraperitoneal after cerebral ischemia 1 hour,The other groups were given the same volume of 0.5% Tween 80.Using Longa 5-point scale to estimate the neurological function;the infarction volume was revealed by TTC stain,the expression levels of PARP-1and AIF were detected by immunehisto-chemistry,The brain morphology were observed by HE stain.All data were expressed as mean ± standard deviation,the comparison among multiple groups used ANOVA,and the comparison between two groups' averages used the t test,with ?<0.05 considered as the significant test standard.Resultneurological function detect were hardly to be found and no infarction was observed in the rat of sham-operation group.The volume of infarction and neurological outcome scores of treatment group [(22.33±4.48)%,(1.50±0.55)]were lower than those of model group[(33.84±3.37)%,(2.67±0.52);P<0.01 respectively],the mean positive cells of the expression of PARP-1 and AIF in model group[respectively(26.17±2.79),(14.67±2.07)] were significantly increased compared with sham-operation group[respectively(5.83±0.75),(4.00±1.79);P<0.01,respectively ];the mean positive cells of the expression of PARP-1 and AIF in treatment group[respectively(16.67±2.66),(9.17±2.93)] were decreased compared with model group[respectively(26.17±2.79),(14.67±2.07),P<0.01 respectively].Conclusionbutylphthalide has neuroprotective effect to cerebral ischemic reperfusion injury,the possible mechanism can be relevant with inhabiting parthanatos.
