To study the process of the synthesis of pazopanib and its two related substances,in order to provide impurity reference substance for the quality control and clinical study of pazopanib.2-methyl-5-nitrobenzenesulfonami--de was used as the starting material to synthesize the target compound 1(Dimer)by sulfonylationand coupling reaction.The target compound 2(Trimer)was synthesizedwith p-chlorotoluene as a starting material by a 7-step reaction consisting of Foucault reaction,amination,reduction,sulfonylation,reduction,deprotection and coupling.Results and conclusion In this study,a metazone-related substance(Dimer)was synthesized and its chemical structure was confirmed by 1H-NMR,13C-NMR and MS.The synthesis method of Dimer is studied.The method shows several advantages,including novelty of route,mild reaction condition,simple operation,high production,high purity of intermediate product and final product.Another related substances(Trimer)was still being synthesized.This study has laid a solid foundation for the extensive preparation of impurity reference compound 1 and the quality control of pazopanib.
