Objective:the purpose of the study was to investigate the mechaism of ERK5 signaling pathway on BMP9 regulate osteogenic/odontogenic differentiation of i SCAP.Methods :Ad-BMP9 was infected into the i SCAP.With the inhibitor of ERK5,BIX02189,the phosphorylated forms of ERK5 protein was assayed by Western blot.Then,the changes of early osteogenic and odontoblast were analyzed by ALP staining.The calcium deposition was examined by Alizarin red staining.At last the expression of osteogenic / odontoblast-associated genes,such as Runx2,OCN,OPN and DMP1,were measured by RT-PCR.Results:BMP9 could up-regulate the phosphorylation of ERK5 in i SCAP.After using BIX02189,which was the inhibitor of ERK5,the phosphorylation of ERK5,the early osteogenic / odontogenic marker ALP and the expression of Runx2,OCN,OPN,DMP1 were significantly inhibited.The calcium deposition was also reduced by BIX02189.Conclusion: ERK5 signaling pathway plays an important role in BMP9-regulated i SCAP osteogenic/odontogenic differentiation.
