OBJECTIVE Propofol is one of the most commonly used intravenous anesthesia inclinical.The repeated application of propofol can induce neuron apoptosis and damage the function learning,memoryand cognitive function.As a perioperative complication hypoxia is easily ignored,which is also an important cause of apoptosis and cognitive dysfunction.In this study,PC12 cells were taken as the research object to investigate the mechanism of propofol exposure leading to PC12 cells apoptosis under hypoxic conditions and provide new ideas for the treatment of postoperative complications and postoperative cognitive dysfunction.METHODS The PC12 cells seeded in culture plates were randomly divided into 6 groups: experiment groups : propofol-hypoxia(PH),propofol-air(PA),propofol-oxygen(PO);control groups: control-hypoxia(CH),control-room air(CA),control-oxygen(CO);The concentration of propofol in all experiment groups is 10?M.In hypoxic groups,the concentrationis of oxygen is 5%;in air groups,the concentration is 21%.What't more,in oxygen groups,the concentration is 35%.After propofol was added,the cells were incubated for 8 h in different oxygen concentration.The cell apoptosis was measured by flow ceytometry,the level of ROS was detected by reactive oxygen species Assay kit and the activity of SOD was evaluated by total superoxide dismutase assay kit with WST-8.RESULTS(1)MTT analysis showed that the proliferation of PC12 cells was significantly inhibited at this concentration of 10?M of propofol for 8h(P < 0.05).(2)Compared with control groups,the apoptosis rate increased significantly in experiment groups(P < 0.05).Compared with the PA and PO,the apoptosis rate was higher in PH group(P < 0.05).(3)Compared with PA,PO and PH,the ROS generation level increased in CA,CO and CH;Compared with PA and PO,the ROS generation level was significantly higher in PH(P < 0.05).(4)Compared with control group,SOD activity increased significantly in experiment groups(P <0.05),Compared with PA and PO,the ROS generation level was significantly higher in PH(P < 0.05).CONCLUSION Propofol caused PC12 cells apoptosis by oxidative stress injury under hypoxic conditions,but the mechanism underlies this effect requires further study.
