Objective To explore the effects of expression of klotho(KL)on the differentiation of RAW264.7 cells into osteoclasts induced by soluble receptor activator of nuclear factor-?B ligand(sRANKL).Methods The RAW264.7 cells were divided into Ad-KL group,Ad-GFP group and control group.The KL transfection into raw 264.7 cells was observed by inverted microscope.The expression levels of KL were detected by RT-PCR and Western blotting method.Tartrate-resistant acid phosphatase(TRAP)staining was applied to observe the osteoclast differentiation and maturation.The expression of TRAP and cathepsin K(CTSK)were assessed by RT-PCR and Western blot.Results Klotho gene was transfected into RAW264.7 cells successfully,Compared with Ad-GFP group and control group,the expression levels of KL mRNA and protein in Ad-KL group were increased(P < 0.01).The TRAP staining results showed that the number and volume of TRAP-positive cells in Ad-KL group was less than those in Ad-GFP group and control group.Compared with Ad-GFP group and control group,the expression levels of TRAP and CTSK mRNA(P <0.01)and proteins(P <0.05)in Ad-KL group were decreased.Conclusion The expression of klotho can inhibit the differentiation of osteoclasts.
