Objective:To detect expression level of SOX15 in colorectal cancer cells and its effect on cell proliferation,apoptosis,migration and invasion.Methods : The expression of SOX15 in Caco2,SW480,HCT116,HT29 colorectal cancer cells and normal colon tissues was detected by RT-PCR.The recombinant plasmid pEGFP-N1-SOX15 and empty vector pEGFP-N1 were transfected into Caco2 and SW480 cells by liposome lipofectamine 2000,and the mRNA and protein expression level of SOX15 in Caco2 and SW480 cells was detected by RT-PCR and Western-blotting respectively.The viability of Caco2 and SW480 cells transfected with SOX15 was tested by cck-8 and the cell proliferation was observed by clony formation assays.The regulation of SOX15 on the ability of cell migration and invasion was used by the Transwell assay.Flow cytometry was applied to analyse the cell apoptosis.Results:The expression levels of mRNA and protein of SOX15 in colorectal cancer cells Caco2,SW480,HT29,HCT116 was significantly lower than that in normal tissues.The expression of mRNA and protein in pEGFP-N1-SOX15 transfected group was obviously higher than that in pEGFP-N1 control group.Contrast to the p EGFP-N1 contral group,pEGFP-N1-SOX15 transfected group was evidently inhibited cell viability,proliferation and the ability of migration and invasion,induced cell apoptosis.Conclusion: Low expression of SOX15 in colorectal cancer cells.SOX15 over-expression could noticeably inhibit the abilities of proliferation,invasion and migration the of colorectal cancer cells,promote cell apoptosis.
