Evaluation Of Antimicrobial Efficacy Of Calcium Hydroxide In Combination With Triple Antibiotiques As Intracanal Medicament Against Enterococcus Faecalis:An In Vivo Comparative Study

Objective The aims of this study were,(i)to evaluate the antibacterial effects of the combination of calcium hydroxide and triple antibiotiques paste as intracanal medicament against E.faecalis,and(ii)to determine the effects of the combination on expression of virulence genes(gel E,cyl A and esp)of E.faecalis biofilms.Methods 65 single rooted premolar teeth were included in the study and chemomechanical preparation of the root canals was performed.E.faecalis infected model was established in vitro.Samples were divided randomly into four groups(n=16): normal saline(NS),calcium hydroxide paste(CH),triple antibiotic paste(TAP),combination TAP with CH(TAP+CH).The colony-forming units were counted at the end of day 3 and day 7.The effects of three medicines on the expression of virulence genes in E.faecalis were evaluated by quantitative polymerase chain reaction.Results 1.The statistical results showed that on day 3 and day 7,the CFU were significantly greater in the NS group than in the study groups(p<0.05).Group TAP+CH and TAP showed significantly less CFU than CH on day 3 and day 7(p<0.05).Group TAP+CH showed significantly less CFU than TAP on day 3(p<0.05).There was no significant difference between the group of TAP+CH and TAP on day 7(p>0.05).2.After treatment,the experimental medicaments all significantly inhibited the expression of virulence genes(gel E,cyl A and esp)than the control group(p<0.05).TAP+CH and TAP both significantly inhibited the expression of virulence genes compared to CH(p<0.05).There was no significant difference between TAP+CH and TAP in the expression of these genes(p>0.05).Conclusions TAP+CH and TAP had more antibacterial effect on E.Faecalis and more effectively inhibited the expression of virulence genes(gel E,cyl A and esp)than CH.TAP+CH and TAP can be considered as an effective alternative intracanal medicament.
