Effect Of Down-regulation Of UHRF1 On Malignant Behaviors In Human Laryngeal Carcinoma Hep-2 Cell Line And The Underlying Mechanisms

Objective : To explore the effect of down-regulation of UHRF1 on proliferation,apoptosis,migration and invasion in human laryngeal carcinoma Hep-2 cell line and the correlative mechanisms.Methods : The experiment was divided in three groups,blank control group,negative control group and siRNA group.Down-regulated the expression of UHRF1 in Hep-2 cell line by UHRF1-si RNA.qRT-PCR(Quantitative Real-time PCR,qRT-PCR)and Western Blot were used to analyze gene expression and protein expression of UHRF1 after down-regulation.The proliferative,invasive and migratory abilities of Hep-2 cells transfected with UHRF1-siRNA were detected by the MTT assay and Transwell chamber assay,respectively.Flow cytometry(FCM)was used to test apoptosis.Westernblot was used to analyze protein expression of Bcl-2,Bax,MMP-2 and MMP-9,respectively.Result:After knockdown of UHRF1-siRNA,the mRNA and protein levels of UHRF1 in Hep-2 cells were significantly reduced.the proliferative abilities of Hep-2 cells were inhibited(p<0.001).The apoptosis rate was(5.47±2.77)%,(3.95±0.66)%,(18.95±1.10)% respectively in the blank control group,negative control group and siRNA group.The apoptosis rate was significantly higher in the siRNA group.The invasive cell number was(213.7±13.05),(221.3±10.26),(97.67±7.51)respectively in the blank control group,negative control group and siRNA group.The migratory cell number was(230.7±5.51),(222.7±11.24),(111.7±7.64)respectively in the blank control group,negative control group and siRNA group.The invasive cell and migratory cell were both decreased in siRNA group(P <0.001).Western Blot showed that Bax was up-regulated(P<0.001),Bcl-2 was down-regulated(P<0.001),MMP-2 and MMP-9 were both down-regulated(P<0.001).Conclusion: Down-regulated the expression of UHRF1 in Hep-2 cell line could induce the apoptosis rate,inhibit the proliferation,reduce the invasive and migratory capability.
