Light licorice from the licorice plant extracts the natural ingredients,has a hundred years of medicinal history,anti-inflammatory and anti-oxidation and treatment of acne and other aspects of a wide range of effects,and long-term as skin care ingredients,so this article to explore the light Licorice is focused on the role and mechanism of treatment of psoriasis.The specific research contents and results are as follows:(1)Isolation and purification of light licorice and structural identification.In this paper,the extraction method was extracted with ethyl acetate for 2h and the ratio of material to liquid was 1:10.The pure liquid was obtained by semi-preparative liquid phase.Confirmation of pure product by high performance liquid chromatography and nuclear magnetic resonance.The HPLC method was established for the determination of the content.(2)Study on the anti-psoriasis activity of light licorice.First of all,Imiquimod emulsion induced mouse psoriasis auricle swelling model and back skin lesion model.Secondly,according to the light licorice in the human skin medication standards set different concentrations,1)The apparent inhibition of the auricle swelling of the mice and the weighed weighing were found to be significantly inhibited by PT(30 g/kg,15 g/kg),and the inhibitory rate was 68.37% and 56.98%.respectively.2)To observe the changes of skin lesions in mice,and to observe the changes of skin lesions in mice by PASI.It was found that PT(30 g/kg,15 g/kg)mice had obvious Suppressing the effect.At last,In view of the above results,HE staining was used to observe the effect of tissue swelling on the swelling and injury of tissue.The results showed that the morphology of the ears and the back skin of the normal group were normal.When using IMQ alone,the skin and the dorsal skin of the mouse were significantly thickened and the infiltration of inflammatory cells increased(cell shrinkage);FK group and PT group were significantly inhibited,and the changes of ear and back skin sections of PT(30 g/kg,15 g/kg)mice were found to be more obvious,including the reduction of ear and skin swelling(Tends to normal group)and the ratio of nucleus to cytoplasm was significantly smaller,the activity is better than the positive drug.(3)Study on the mechanism of light licorice against psoriasis.Collect the above mouse psoriasis tissue samples,extract proteins and mRNA.The expression of each target factor in tissues was detected by RTF-QPCR and IHC.The results showed that the expression levels of P65,IFN-?,IL-17 A,IL-6,COX-2,IL-22,IL-23,CD4 and CD8? in the skin of mice were significantly increased when used by IMQ alone,The PT groups had a certain degree of inhibitory effect on these inflammatory mediators,and the high concentration of 30 g / kg had the strongest inhibitory effect and was better than the positive drug.It can be seen that the light of licorice is by inhibiting the IL-23/IL17 inflammatory response axis and more inflammatory factors show anti-psoriasis effect.In summary,this paper from the light of licorice extract detection and anti-psoriasis activity and pathology research,light licorice for the application of psoriasis to provide theoretical and experimental basis.
