Objective: Gastric cancer is one of the most common gastrointestinal tumors,the incidence and mortality of it is in the first place.Invasion and migration of gastric cancer which are the biological characteristics are the leading cause of death and worse prognosis of patients,closely related to the tumor microenvironment.TSPAN9 is one of the transmembrane 4 superfamily members,the research about it is limited.Our study intends to explore the effect of TSPAN9 on gastric cancer SGC7901 cells in the acidic microenvironment about invasion and migration.Methods: With the synthetic TSPAN9 gene was inserted into vector to obtain the plasmid,and then transfected into TOP10 clone strains to get more.Next,we defined SGC7901/TSPAN9 as transfection group,while SGC7901 as blank group and SGC7901/NC as control group.The establishment of acidic microenvironment was achieved by adding 3% HCL and the final PH adjusted to 7.4 or 6.5,culturing cells in standard incubation conditions.Western Blot and q RT-PCR were used to verify the overexpression of TSPAN9 at RNA and protein level.The effects of TSPAN9 on invasion and migration in acidic microenvironment were observed by Wound healing and Transwell assay.At the same time,Western blot was used to detect the expression of u PA and MMP-9.Results: SGC7901/TSPAN9 group in the acidic microenvironment,the expression of TSPAN9 is higher in RNA and protein level.The tumor acidic microenvironment promotes the invasion and migration of gastric cancer,P<0.05.Compared to the three groups,we can find that 6.5-SGC7901/TSPAN9 group can significantly promote the expression of u PA,MMP-9,promoting the invasion and migration,P<0.05.Conclusions: Tumor acidic microenvironment promotes the expression of TSPAN9 on m RNA and protein level,and promotes the invasion and metastasis of gastric cancer cells.Tspan9 promotes migration and invasion of human gastric cancer SGC7901 cells in the acidic microenvironment,providing a new target for the gastric cancer in acidic microenvironment.
