The Research Of Serum Level Of Orexin A And Expression Of Its Receptor In Patients With Gastric Cancer

Objective To investigate the different serum level of Orexin A(OXA)in patients with gastric cancer and healthy people by enzyme-linked immunosorbent assay(ELISA),and detect the expression of orexin-1 receptor(OX1R)by immunohistochemical staining.Exploring the role of orexin A in the occurence and progression of gastric cancer.Methods To detect the serum level of Orexin A(OXA)of 50 patients with gastric cancer and 30 healthy subjects by enzyme-linked immunosorbent assay(ELISA).The distribution of orexin receptor 1(OX1R)in gastric carcinoma and normal gastric mucosa was detected by immunohistochemical staining.analyzed statistically.And statistical analysis was performed.Results(1)In the two serum groups,the levels of Orexin A in patients with gastric cancer are higher than those in healthy people,the difference is statistically significant(P < 0.01).(2)We divided the two serum groups into two groups according to age,respectively,namely“?65 years old”and“>65 years old”,there are both no obvious difference between the level of Orexin A and the age in these two groups(t = 0.59?t=0.68,P> 0.05).(3)The serum samples of gastric cancer were divided into undifferentiated gastric cancer,poorly differentiated carcinoma,moderately differentiated carcinoma and well differentiated adenocarcinoma,there is no obvious difference between the level of Orexin A and the stage of adenocarcinoma(F=0.05,P>0.05).(4)In normal gastric mucosal tissue,the positiveexpression rate of Orexin receptor type 1 is 60% and the positive average staining score is(3.67 ± 1.80);The positive expression rate of Orexin receptor type 1 in gastric cancer tissue is 94.7% and the positive average staining score is(7.53 ±3.20).Compared with normal gastric mucosal tissue,the positive expression rate of Orexin receptor type 1(P<0.01)and the positive average staining score(P<0.01)in the gastric cancer tissue are significantly higher.(5)The gastric cancer tissues were divided into differentiated gastric cancer and undifferentiated gastric cancer according to the pathological stage.The average staining score of Orexin receptor type 1 positive expression in differentiated gastric cancer tissue is(6.86 ±2.87),the average staining score of undifferentiated gastric cancer was(9.88 ±2.41).Compared with differentiated gastric cancer,the average staining score of undifferentiated gastric cancer was significantly higher(P <0.05),and the detected Orexin receptor type 1 mainly locate in the cytoplasm and membrane of the cancer cells,light yellow to brown granules.Conclusion The level of orexin A in the serum of patients with gastric cancer increased,and the expression of OX1 R in gastric cancer tissues increased,suggesting that the orexin system plays an active role in the development and progression of gastric cancer.
