Objective To evalute the role of Sufentanil preconditioning on MAPK signal pathway and expression of connexin 43Methods Primary cardiomyocytes were cultured for 5 days were randomly divided into 13 groups:control group(group C),hypoxia / reoxygenation(H / R group),morphine preconditioning group(group M),sufentanil preconditioning group(group S),ERK inhibitor group(group IE),JNK inhibitor group(group IJ),p38 MAPK inhibitor group(group IP),ERK inhibitor+morphine preconditioning group(group MEB),ERK,inhibitor +sufentani preconditioning group(group SEB),JNK inhibitor+morphine preconditioning group(group MJB),JNK inhibitor+sufentanil preconditioning group(group SJB),p38 MAPK inhibitor +morphine preconditioning group(group MPB),p38 MAPK inhibitor + sufentanil preconditioning group(group SPB).The cells were exposed to hypoxia solution saturated with mixture of 95% N2+5% CO2 for 2 h,and cultured in the normal atmosphere for 1h.Cx43 changes in each group were measured average optical density(AOD)by confocal microscopy;the Cx43 protein expression and Cx43-Ser368 sites phosphorylation calculated by Western Blot.Results: Compared with group C,the rest group of Cx43,p-Cx43Ser368 protein and AOD expression reduced(P(27)0.05);Compared with group H/R,Cx43,p-Cx43Ser368 protein and AOD expression were significantly increased in group M,group S,group MEB,group SEB,group MJB,group SJB,groupMPB,group SPB(P(27)0.05),group IE,group IJ,group IP showed no significant difference(P>0.05);compared with group M,Cx43,p-Cx43Ser368 protein and AOD expression decreased in group IE,group IJ,group IP,group MEB(P(27)0.05),group S,group MJB,group MPB were showed no significant difference(P>0.05);compared with group S,Cx43,p-Cx43Ser368 protein and AOD expression decreased in group IE,group IJ,group IP,group SEB(P(27)0.05),group SJB,group SPB were no significant difference(P> 0.05).Conclusion: Sufentanil pretreatment can produce similar protective effect on Cx43 with morphine,enhance the level of Cx43-Ser368 sites phosphorylation;This effect is associated with ERK signal pathway,also may be related to JNK,p38 MAPK signal pathway.
