Objective: Hydatid disease(also called Echinococcosis)caused by the larvae of echinococcus granulosus living in human and animals,was a parasitic zoonosis that causes seriously harmful for human health and the development of stock farmin.At present,hydatid disease patients were about 60 ~ 300 million in our country,surgery was the best treatment,but postoperative recurrence,and adjuvant therapy was few in category and species,Poor efficacy,adverse reactions obviously.The purpose of this study was to develope new drugs for the treatment of hydatid disease,aimed to remove residual echinococcus,diminish baneful adverse reaction,to achieve the goal of improving immune function,low toxicity,improving patient compliance by using the characteristic of chinese and tibetan medicine of multi component,multi target,rich in resources.Methods:70 healthy kunming mice,weighing 18-20 g,were randomly divided into 2 groups(experimental group and negative control group),experiment group 60,negative control group of 10,sex in half.Each of mouse in experimental group was injected intraperitoneally with 0.3ml fully mixing protoscoleces suspension(4500/ only),negative control group was injected with normal saline 0.3ml,feeding 3 months at a temperature of 18-22?,humidity 50-60% environment.60 mice in the experimental group were randomly divided into 6 groups once more(alkaloid low,middle and high dose group,albendazole medication group,albendazole + alkaloid group,positive control group),each with 10,sex in half.The mouse in low,medium and high dose groups were given alkaloid of 8mg/kg.d,12mg/kg.d,16mg/kg.d by the method of gavage administration respectively.The albendazole group was administered at a dose of 20 mg / kg.d,and the mixed drug group(albendazole +alkaloid group)was administered at 10+ 8 mg / kg.d,and the positive control group was given saline.Continuous administration for 30 days.Each mouse was weighted,taking blood by enucleating eyeball after disinfection,3000r/min centrifugation,collecting and keeping serum at 4 to be measured.After the mice were sacrificed,?the thymus,spleen,and cyst were weighed.Calculating the thymus index(thymus index = thymus mass / body weight *100%)and spleen index(spleen index = spleen mass / body weight *100%),(the cyst inhibition rate=(Cyst weight of positive control group-Cyst weight of drug group)/Cyst weight of positive control group*100%).Drawing standard curve of each immunological factor according instructions,determining the absorbance of each index,and calculating the content of each indicator according to the standard curve equation.Results: Thymus index analysis showed a decreasing trend after mice infected with hydatid disease,and recovery effect obviously after the treatment of alkaloids from Sophora moorcroftiana,The effect of albendazole treatment group,albendazole and alkaloid mixture group were less than that of the alkaloid group,but there was no significant difference between each groups(P>0.05).When the mice were infected with echinococcosis,the spleen had a compensatory increase,and spleen increased the most obvious in combination therapy group,But the difference was not statistically significant(P>0.05).The analysis of hydatid cyst weight show alkaloids from Sophora moorcroftiana and albendazole could inhibit the growth of cysts,and the inhibition rate was the most obvious in the albendazole group and the combination group,but there was no significant difference between each groups(P>0.05).The levels of aspartate aminotransferase(AST)and alanine aminotransferase(ALT)were increased after infected with echinococcosis,and the difference of aspartate transferase(AST)was statistically significant(P <0.05).After treatment with drugs,the aspartate transferase(AST)and alanine aminotransferase(ALT)in the albendazole group were increased,while the other groups were decreased,especially in the mixed group,but there were no statistically significant between each group(P> 0.05).The level of IL-2 was relatively reduced after infected,and the difference was statistically significant(P <0.05);IL-2 levels were recovered after administration,and there were statistically significant compared with the positive control group(P <0.05);The recovery of alkaloid group was lower than that of albendazole group,and the difference was statistically significant(P <0.05);The level of recovery after drug combination was increased,and there was no statistically significant compared with albendazole group(P <0.05).IL-6 levels were elevated after infected with echinococcosis,and recovered after treatment,the difference was statistically significant compared with the positive control group(P <0.05),but no tatistically significant between each other(P>0.05).IL-10 levels were reduced after infected and recovered after treatment,but there was no significant difference compared with the positive control group(P> 0.05).Ig E analysis showed that Ig E increased after infection with echinococcosis in mice,and the difference was statistically significant(P <0.05).After treatment,Ig E levels were recovered,and there was significant difference compared with the positive control group(P <0.05).TNF-? analysis showed no significant change before and after infection and treatment,the difference was not statistically significant(P> 0.05).Conclusions: Ig E?IL-2?IL-6 could be used as evaluation index in the production of hydatid disease animal model and dlinical drug therapy for the immune indicators can be changed before and after infection with echinococcosis and medical treatement.The characteristics of alkaloids from Sophora moorcroftiana of Multi-component,multi-target,low toxicity,improve immune function,improve patient compliance contributed to the treatment of hydatid disease.it could not only improve the efficacy,but also helped to reduce its liver toxicity,protected animal with liver function,especially combined with albendazole.So the combined medication was the focus on the treatment of hydatid disease henceforth.
