Objective:To find the best treatment time and the best dose of hydrogen sulfide postprocessing on transient global cerebral ischemia.In addition,Kv4.2 and VEGF play an important role in neuroprotective mechanisms,but their role in this model and their relationship with H2 S remain to be investigated.We decide to find the relationship between Kv4.2,VEGF and hydrogen sulfide.Methods:Male Wistar rats were selected,and their body weight was 200-280 g.Transient global cerebral ischemia model was made by four vessel occlusion.After different hydrogen sulfide treatment time and different hydrogen sulfide treatment.Nissl staining was used to count the number of cells in CA1 area of hippocampus in rats,and the optimal post-treatment time and dose were determined.The expression of Kv4.2 and VEGF in different groups were observed by Western blot.Results:(1)Hydrogen sulfide postprocessing can protect the hippocampal CA1 neurons after transient cerebral ischemia;(2)Hydrogen sulfide postprocessing was effective only when applied 12-48 hours after transient global ischemia.;(3)Different postprocessing times have different effects;(4)Compaired with t GCI group,12 h24umol/kg group and 12h48umol/kg group have a significant protective effect(p < 0.05),and 48umol/kg group have more survive cells than24umol/kg group(P < 0.05);(5)Postprocessing applied 24 hours after transient global ischemia,24umol/kg group and 48umol/kg group have significantly protective effect(P < 0.05),but the protective effect between the two groups had no significant difference(P>0.05);(6)Postprocessing applied 48 hours after transient global ischemia,only24umol/kg group has a protective effect(P < 0.05);(7)When hydrogen sulfide was given at 24umol/kg,different postprocessing time have different protective effects,the dose of 24umol/kg group has the strongest protective effect(P < 0.05);(8)In hydrogen sulfide given 48umol/kg postprocessing,24 h group has the strongestprotective effect(P < 0.05);(9)144umol group in ischemia reperfusion for 12 h group,24 h group and 48 h group were found no protective effect;(10)The expression of Kv4.2 in 38 hours after ischemia decreased compared with sham operation group(P< 0.05),but given the hydrogen sulfide group was expressed higher than ischemia group(P < 0.05),but no more than sham operation group(P < 0.05);(11)Compared with the sham group,the VEGF expressionr of ischemia group was significantly increased(P < 0.05),given the sulfide group was increasing obviously than ischmia group(P < 0.05).Conclusion:(1)In the cerebral missing 12-48 hours after giving small measurement of hydrogen sulfide postprocessing can play a neuroprotective effect,the same measurement administration protection at different time have different effects;(2)Hydrogen sulfide may inhibite the decrease of Kv4.2 and upregulation of VEGF achieve neuroprotection.
