Objective: Endometrial carcinoma is an epithelial malignant tumor of the endometrium.The proportion is about 8% in female malignant tumors and 20%~30% in malignant tumors of female genital system.The number of new cases of endometrial cancer in China is about 63400 with an estimated mortality rate of about 21.8%,and new cases is on the rise in 2015.Endometrial carcinoma became the top second gynecological cancer following breast cancer in the United States with a new case of about 60050 people in 2016.SHNG5(small nucleolar RNA host gene 5,SNHG5)is the mature cut of U50HG' exon.U50 HG is the host gene of the small nucleolar RNA(snoRNA)U50 and U50'.Researchers have found that SNHG5 was reduced in gastric cancer,and down-regulation of SNHG5 suppressed proliferation,migration,and invasion of gastric cells in vivo and vitro.SNHG5 might affect acetylation by trapping MTA2 in the cytosol,thereby interfering with the formation of the nucleosome remodeling and histone deacetylation complex.To investigate the expression of Long non-coding RNA SHNG5 in endometrial carcinoma(EC)and its relationship with clinicopathological features and patients survival.Methods:1 A total of 108 fresh endometrioid carcinoma tissues were obtained from patients who underwent surgery at Department of Gynecology,the Fourth Hospital of Hebei Medical University from January 2010 to December 2012,of which 91 cases of ?type endometrial carcinoma,17 cases ? type endometrial carcinoma(including 11 cases serous adenocarcinoma,5 cases clear cell carcinoma and 1 case undifferentiated carcinoma).According to the 2009 International Obstetrics and Gynecology staging(FIGO 2009),there were 85 patients in stage ? and stage ?,23 patients with stage ? and stage ?,and 32 cases of G1 grade,59 of G2 grade,17 cases of grade G3 according to the pathological grade.All patients with endometrial carcinoma had not been treated with radiotherapy,chemotherapy and hormone therapy before operation.Patients were aged between 35 and 72 years with an average age of 55±9 years.66 normal endometrial carcinoma tissues were abtained from patients who underwent hysterectomy in the same period and been pathological examination confirmed for gynecological benign disease.Age of patients were 42 to 62 years,with an average age of 52±12 years.2 Total RNA from tissues was extracted using the TRIzol reagent according to the manufacturer's protocol.The expression of SNHG5 in normal endometrial tissue and endometrial carcinoma was detected by Real-time PCR(qPCR).3 The EC group was devided into two groups according to the median level of SNHG5 in EC tissues.The correlation between SNHG5 expression in EC and cilinicopathologic features was analysed.4 Statistical analyses were performed with SPSS 13.0 statistical software.The comparison of SNHG5 expression between two groups was performed using the Wilcoxon rank sum test,expressed by M(QR);The association of SNHG5 expression with clinicopathological features was evaluated using Pearson's c2 test.Survival curves were plotted by the Kaplan-Meier method,and the log-rank test was used to assess differences between stratified survival groups using the median value as the cutoff.A two-sided P value of P<0.05 was considered to be statistically significant.Result:1 The expression of SNHG5 in EC.Real-time PCR results showed that he expression of SNHG5 in EC were lower than normal tissues(Median:1.62,1.88,Interquartile Range:0.86,0.91)(P<0.05).2 The relationship between SNHG5 expression and clinicopathological features of EC patients.There were no significant differences between SNHG5 lower expression group and SNHG5 higher expression group in age,histological grade,depth of myometrial invasion and negative rate of vascular invasion,(P<0.05).But the expression of SNHG5 in patients with lymph node metastasis positive was lower than patients with lymph node metastasis negatitive(P<0.05).3 The relationship between SNHG5 expression and overall survival time of EC patients.The patients with lower expression of SHHG5 had significantly lower survival time after operation(41.78±12.64 months)than patients with higher expression of SHHG5(53.70±9.73 months)(P<0.05)?Conclusion: Expression level of SNHG5 in endometrial carcinoma tissues is significantly decreased and low expression of SNHG5 is related to the poor prognosis of endometrial cancer patients.These results suggest that SNHG5 may inhibet of endometrial cancer progressionbe an important molecular marker for the prognosis of endometrial carcinoma.
