Objective: Lung cancer is one of the most serious malignant tumors which threaten the health and life of human.In the recent 50 years,the mortality rate and the death rate were both significantly increased.Though its pathogenesis was not completely clear,the reasons could be roughly divided into several aspects.For example: tobacco exposure,occupation and environmental factors,air pollution,genetic factors and so on.With the gradual and in-depth understanding of the disease,more and more attention had been paid to the study on gene molecular level.To predict overall survival of patients with non-small-cell lung cancer and provide a basis for individualized treatment of non-small-cell lung cancer,some genes which were related to the prognosis of non-small-cell lung cancer were chosen by using high throughput screen of gene technology.Therefore,the genetic testing was gradually valued.Relevant research showed that NF1?HMMR had different expressions in the patients with non-small-cell lung cancer.The expressions of NF1?HMMR in nucleic acid which was extracted from the blood and tissue of the patients with non-small-cell lung cancer were tested by HRM experiment operation in the laboratory.In the article,we aimed at exploring the feasibility of using the experiment operation to detect the NF1 and HMMR genes by the lung cancer tissue samples(the lung cancer tissue wax blocks)and the blood of patients with non-small-cell lung cancer.Methods:1 30 blood specimens(blood specimen group)and 18 lung cancer tissue wax blocks(tissue specimen group)were collected randomly from 204 patients with non-small-cell lung cancer who were treated by thoracic surgery in the fourth hospital of Hebei Medical University from September 2010 to September 2015.Before surgical treatment,all patients did not accept chemotherapy or radiation therapy.Besides,all of them did not have other cancers or some angiogenesis related diseases such as endometriosis;respiratory distress syndrome;chronic heart failure;chronic respiratory failure,diabetes and so on.All of them are the Han nationality and there was no history of long-term exposure to clear carcinogenic factors.Pathological types of tumors were confirmed by pathological report of surgical resection specimens from the Fourth Affiliated Hospital of Hebei Medical University.2 As the template of PCR reaction,nucleic acid was extracted from the blood of the patients with non-small-cell lung cancer by the technology of human blood MINI BLOOD DNA in the laboratory.The expressions of NF1 gene and HMMR gene were detected by HRM experiment operation and compared the product which were amplified by PCR with standard NF1?HMMR genes.Then we analyzed the results.3 As the template of PCR reaction,nucleic acid was extracted from the lung cancer tissue in the laboratory.The expressions of NF1 gene and HMMR gene were detected by HRM experiment operation and compared the product which 19.0 statistics software,we explored the reliability of the experiment operation to detect the expression of the two genes(NF1?HMMR)in the blood and lung cancer tissues of patients with non-small-cell lung cancer.Results:1 The expressions of NF1 gene and HMMR gene in blood samples were detected by the experiment operation.The DNA extracted from the blood had been transcribed to RNA.The quality of RNA was poor and the result was meaningless.There were no positive test results.Tissue DNA or tissue wax block is recommended;2 It is feasible to detect the expressions of NF1 and HMMR gene in tissue sections of lung cancer by experiment operation.The positive rate of gene expression in lung cancer tissue was 44.4%.The positive rate of NF1 expression was 33.3%(6/18),and the positive rate of HMMR gene was 38.9%(7/18).The experimental results showed that there were significant differences between blood specimen group and tissue specimen group(P<0.05);3 The positive expression of NF1 gene in non-small-cell lung cancer patients was correlated with lung cancer staging(lymph node metastasis and tumor size P<0.05).Conclusions:1 In this paper,it was poor to detect the expression of NF1 gene and HMMR gene in the nucleic acid which was extracted from the blood of the patients with non-small-cell lung cancer by using HRM experiment operation.The positive detection rate was 0.RNA could not be detected effectively when it was transcribed from DNA in blood samples,and it was closely related to the little content of nucleic acid which was extracted from blood.2 It is feasible to detect the NF1 gene and HMMR gene in non-small-cell lung cancer tissue.By comparing the blood specimen group with the tissue specimen group,it is more suitable to detect the lung cancer tissue as the method of testing NF1 and HMMR genes.3 According to the analysis of the results of small sample and the basic data of patients,the high expression of NF1 had a positive effect on the prognosis of patients.This result was consistent with the conclusion that NF1 gene was a protective gene.
