Nowadays,the lung cancer is one of the most common malignant tumor in the world and the survival rate is low.Although the treatments,in form of the surgery,chemotherapy,radiation therapy etc.,are used,the mortality of lung cancer can,t be significantly reduced.The rapid development of molecular biology and immune technology provide a more adequate theoretical basis and practical experience for the lung cancer immunotherapy and are playing a more and more significant role in the comprehensive treatment of lung cancer.Therefore,they are expected to become one of the cue for lung cancer.The immune response is a continuous and complex process.With the further study on the immune regulation mechanism,the important role of cytokines in anti-tumor immune response is gradually taken seriously.The research on cytokines and lymphocytes that secrete cytokines will become the focus in the tumor biological treatment,which has a good application prospect.Tumor sells break the balance between immune cells and cytokines in peripheral blood.Recently the new auxiliary T cell Th17 whose feature is to secret IL-17 is identified.Purposes:Apply CBA to detect the expression level of the cytokines Th1,Th2,Th17 in peripheral blood of patients with lung cancer and to research the significance of the change of cytokines in the peripheral blood of cancer immune microenvironment.Methods:1 The selection of the research objectConsidering the 50 diagnosed patients with lung cancer in the affiliated hospital of hebei university as a observation group and the healthy people of physical examination as a control group.The people in control group have no other diseases.2 The collection of serum samplesBlood the patients of observation group before and after 2 cycles of chemotherapy and abstract respectively from two groups of patients with peripheral blood 5ml on empty stomach in the morning.Get the supernatant blood and tag it.3 The detection of effect factorsDetect respectively the concentrations of cytokines Th1?TNF-?,IFN-?,IL-2?,TH2?IL-4,IL-6,IL10?,TH17?IL-17?by using CBA.Results1 The cytokines' changes in the observation group and the control group before chemotherapy:Compared with the control group,the cytokines Th2?IL-4,IL-6,IL-10?,Th17?IL-17?in the observation group obviously increase?P<0.05?,while Th1?TNF-?,IFN-?,IL-2?obviously decrease?P<0.05?.2 The cytokines' changes before and after chemotherapyCompared with before chemotherapy,in the observation group the cytokines Th2?IL-4?,Th17?IL-17?after chemotherapy decrease obviously,while Th2?IL-6,IL-10?has a downward trend.But no statistical differences?P>0.05?;Th1?TNF-?,IFN-?,IL-2?increase obviously?P<0.05?.3 The observation group and the control group after chemotherapyCompared with the control group,in the observation group the cytokinesTh2?IL-4,IL-6,IL-10?,Th17?IL-17?obviouslyincrease?P<0.05?;Th1?T NF-?,IFN-?,IL-2?obviously decrease?P<0.05?.But the range of changing decrease after chemotherapy.4 In the healthy people,IL-17 is not obviously related to the cytokines Th1 and,Th2?P>0.05?;In patients with lung cancer IL-17 is obviously related to the cytokines Th1?IFN-?,IL-2?and Th2?IL-6,IL-10??P<0.05?.Conclusions:1 The cytokines Th1 in peripheral blood of patients with lung cancer were lower while the cytokines Th2 and Th17 were higher,suggesting that maybe the cause of immune function of patients with lung cancer drops and that it is related to the lung cancer's occurrence and development.2 Maybe the effective chemotherapy can improve the immune function of patients with lung cancer.
