The human brain glioma is the most common intracranial malignant tumor,at present the treatment of glioma is mainly based on surgery,and combined with radiation and chemotherapy.But those treatment means had poor effects,high disability rate and poor prognosis.Molecular targeted therapy of malignant tumor is the current research hotspot.The occurrence and development of glioma is the result of joint action of many factors.The effect of VEGF,ILK and CD151 on promoting tumor angiogenesis may be related to the growth and invasion of the human brain glioma.This research is through the method of immunohistochemistry to detect the expression of VEGF,ILK,and CD151 in the different pathological levels of human brain glioma and normal brain tissue,then statistics and analyzes the correlation among their expression,to study the possible effect mechanisms of the three proteins.We may found some new targets for molecular targeted therapy of combination anti-angiogenesis on human brain glioma.Objective:Detect the expression of VEGF protein,ILK protein,and CD151 protein in human brain glioma and normal brain tissue and analyze the expression differences and correlation among different groups,to explore the role of the three proteins in gliomas and the relationship between their expression levels and the pathologic grade of human brain glioma.Discuss the mechanisms of three proteins in the angiogenesis of human brain glioma and the relationship with the growth and invasion of the human brain glioma,to provide new theoretical basis for the anti-angiogenesis therapy of human brain glioma.Methods:We collected 75 cases pathological block of human brain glioma tissues,Glioma tissues were graded by the WHO classification criterion of centralnervous system tumors in 2007: ? level 8 cases,? level 24 cases,?level17 cases,? level 26 cases.We defined ?and ? as the low grade group,? and ? as the high grade group.We also collected 15 cases pathological block of normal brain tissues as the control group,and those tissues were collected from those serious craniocerebral trauma patients who accepted operation after craniotomy for cerebral decompression.Immunohistochemistry was used to detect the expression of VEGF,ILK,and CD151 in three groups.We analyze the relationship between the expression levels with glioma malignant degree and the correlation among these expression levels in glioma.The experimental results were analyzed by using SPSS 19.0 statistical software.Categorical data using ?2 test,correlation using Spearman relevance measurement,?=0.05 using as the test standard,define P<0.05 as the differences having statistical significance.Results:1 The expression levels of VEGF,ILK and CD151 in three groupsVEGF protein expression mainly localized in the cytoplasm and some localized in the vascular endothelial cell cytoplasm,and the expression levels of VEGF protein in the high grade group,the low grade group and the control group were 74.42%(32/43),46.88%(15/32),6.67%(1/15);ILK protein expression mainly localized in the cytoplasm and cell membrane,and the expression levels of ILK protein in the high grade group,the low grade group and the control group were 83.72%(36/43),59.38%(19/32),20.00%(36/43);CD151 protein expression mainly localized in the cytoplasm,and the expression levels of CD151 protein in the high grade group,the low grade group and the control group were 81.40%(35/43),56.25%(18/32),13.33%(2/15).2 The respective expression differences of VEGF,ILK and CD151 in three groupsPositive expression rates of VEGF,ILK and CD151 in glioma group were all significantly higher than in the normal brain tissue,the expression difference was statistically significant(P<0.05).The expression levels ofVEGF,ILK and CD151 in the high grade group is higher than in the low grade group,the expression difference was statistically significant(P<0.05),and the positive expression rates of the three proteins were increasing with the increasing of glioma pathological grading.3 The expression of VEGF,ILK and CD151 in gloma and their correlationIn glioma tissues,the expression of VEGF and ILK protein were positively correlated(? 2=5.989,r=0.283,P<0.05),the expression of ILK and CD151 protein also positively correlated(? 2=3.942,r=0.229,P<0.05),the expression of VEGF and CD151 protein were positively correlated(?2=5.629,r=0.274,P<0.05).The expression of VEGF,ILK and CD151 in glioma tissues were positively correlated between each other,and the correlation coefficient of VEGF and ILK was the highest,the correlation coefficient of VEGF and CD151 was the lowest.Conclusions:The positive expression rates of VEGF and ILK CD151 in glioma tissues is significantly higher than that in normal brain tissues.The expression levels of VEGF and ILK CD151 in high grade glioma was different with that in low grade glioma,and the positive expression rates increased with the increased of glioma pathology levels.The expression of VEGF,ILK and CD151 in glioma tissues were positively correlated between each other,they may work together in the occurrence and development of glioma blood vessels,and promote the growth and invasion of glioma.Therefore,if through interdicting the expression,transcription of the three proteins or inhibiting the effect of their proteins may inhibit the angiogenesis of glioma,and then interdict the pathway of supply and cancer metastasis.Finally inhibit the growth and infiltrating invasion of glioma.VEGF,ILK and CD151 expression in the tissue of gliomas is closely relative to between two and three may and Therefore,through joint,blocking the expression of transcription or inhibit the role of the protein,or inhibiting the production of glioma tumor blood essels,which block the nutrient supply and cancer metastasis pathway,Speculatedthat this may be as a potential molecular targets of glioma angiogenesis therapy,may become the new direction for the treatment of gliomas.We suspect that those may be potential molecular targets of glioma angiogenesis therapy,and this may become the new direction for the treatment of gliomas.
