Cancer has been one of the hugest challenges faced by the human being in recent years;millions of people died of cancer every year in the world.The cervical cancer is a common one of the most malignant tumors in female,the incidence of which is very high in women from the developing countries.This study is to explore the effect of curcumin on growth and proliferation of human cervical cancer He La cell,aiming at providing the evidence and foundation for elucidating the mechanism of curcumin in tumor therapy.He La cell was treated with different concentrations of curcumin,0 μmol·L-1,5 μmol·L-1,10 μmol·L-1,20 μmol·L-1,in vitro.MTT,Brdu staining and plate cloning experiments were used to study the effect of curcumin on growth of He La cell.The results showed that curcumin had an inhibitory effect on the growth of He La cell in a dose-dependent manner.Immune fluorescence confocal experiment was used to study the localization of curcumin and UBF in the cell.The results showed curcumin was co-localized in the nucleolus with UBF.The real-time polymerase chain reaction experiment was used to detect the nucleolus transcription of He La cell.The results showed curcumin could down-regulate the level of nucleolus transcription.Western blot was used to study the level of phosphorylation of UBF.The results show curcumin could down-regulate the level of phosphorylation of UBF.This study demonstrated that curcumin could inhibit the growth of He La cell in a dose-dependent manner at a cellular level.Moreover,curcumin could down-regulate the level of nucleolus transcription and the level of phosphorylation of UBF.Hence,we inferred that curcumin could inhibit the growth and proliferation of He La cell through the down-regulation of the level of phosphorylation of UBF and the level of r DNA transcription.Considering the complexity of cell metabolism and the wide biologicl effects of curcumin,the transcription factor UBF may be a target of curcumin on cancer therapy;further work needs to be done to explore the molecular mechanism.
