Objective: Using immunohistochemical method to detect the expression of epithelial markers(CK7,E-cadherin)and mesenchymal markers(Vimentin,N-cadherin)in biliary epithelial cells of human Hepatolithiasis,to explore Whether the EMT occur in biliary epithelial cells.Whether TLR4 participate the process of EMT;The clinical significance of theses markers among their expression.Methods: Collected the liver tissues of 32 patients with intrahepatic biliary stones as observation group,and the liver tissue of 14 cases with hepatic cavernous hemangioma as control group,Retrospectively.Used the immunohistochemical methods to detect the expression of CK7,E-cacherin,Vimentin,N-cadherin and TLR4 in the biliary epithelial cells,and the interrelation among the expressions of these markers will be analyzed.Result:1.Immunohistochemical staining results showed that CK7 and E-Cadherin were strong positive expression in bile duct epithelial cells of the control group,the expression decreased significantly in the observation group,even missing;Vimentin and N-cadherin were almost no expression in bile duct epithelial cells of the control group,and were weakly positive expression in the observation group;TLR4 was weakly positive expression in the control group,but was significantly enhanced expression in the observation group.Among the observation and control group,the expression of CK7,E-Cadherin,Vimentin,N-Cadherin and TLR4 were all significantly different,P<0.05.2.The expression of CK7 was significantly correlated with Vimentin and N-cadherin,a negative correlation among them(r=-0.793,-0.680).The expression of E-cadherin was significantly correlated with Vimentin and N-cadherin,a negative correlation among them(r=-0.812,-0.660).3.The expression of CK7 and E-cadherin were significantly correlated with the pathologic classification of Hepaticlithiasis,there are significant differences and positive correlation between ? and ? type(P=0.027?0.027,r=-0.502?-0.502),but ? type had no significant difference with ? and ? type;Likewise,the expression of Vimentin and N-cadherin were significantly correlated with the pathologic classification of Hepaticlithiasis,there are also significant differences and positive correlation between ?and ? type(P=0.014?0.037,r=0.550?0.479),but ? type had no significant difference with? and ? type.The expression of all makers have no significant differences with the gender,age and distribution of stones in Hepaticlithiasis,P>0.05.Conclusions:1.Under the observation of this experiment,the biliary epithelial cells may undergo the process of EMT in pathological changes of human Hepatolithiasis.2.The EMT of biliary epithelial cells was positively correlated with the degree of fibrosis of biliary in Hepatolithiasis.3.TLR4 may participate the process of EMT in biliary epithelial cells of human Hepatolithiasis,...
