Objective:On the based of Polygonum cuspidatum cream,to optimize the microemulsion gels local drug delivery system to on the design and evaluation research,laid the foundation for the improvement of the anti-inflammatory ointment type of Polygonum cuspid.Methods: 1.To study the preparation technology of Polygonum cuspidatum extract:As the evaluation index to polydatin and emodin and extraction rate of extractum,select the ethanol concentration,extraction temperature,extraction time and material liquid ratio were investigated,using ethanol reflux extraction process of uniform design optimization of Polygonum cuspidatum;emodin rate,transfer rate and drying time as the evaluation index to polygonin transfer study the factors to concentrate drying temperature and concentration drying method,the concentrated extract of Polygonum cuspidatum,drying conditions were studied;Study on Preparation and testing of extracts.2.To study the prescription and process of Polygonum cuspidatum microemulsion gel:Study on the commonly used emulsifier,oil phase solubility in Polygonum cuspidatum extract,and by single factor experiment and Box-behnken experiment design of emulsifier,emulsifier types,Polygonum cuspidatum in microemulsion help oil phase and optimize the dosage and selection method of substrate concentration for preparation of microemulsion gel were investigated by gel material of suitable material.3.Evaluation of in vitro absorption of Polygonum cuspidatum microemulsion and Polygonum cuspidatum microemulsion gel:In vitro transdermal absorption experimentswere carried out to investigate the transdermal speed and retention of the prepared Polygonum cuspidatum microemulsion,Polygonum cuspidatum microemulsion gel and the anti-inflammatory cream of Polygonum cuspidatum.4.Local pharmacokinetics study of Polygonum cuspidatum microemulsion gel:The local skin pharmacokinetics of the prepared Polygonum cuspidatum microemulsion gel and the anti-inflammatory cream of Polygonum cuspidatum were studied.The retention of polydatin and emodin in the skin at different time points was investigated and the pharmacokinetic parameters were calculated.5.Preliminary pharmacodynamic study of Polygonum cuspidatum microemulsion gel:In the experiment of anti-inflammatory,anti-inflammatory ointment,microemulsion gel of Polygonum cuspidatum and model group with significant difference(P < 0.01),analgesic experiments of Polygonum cuspidatum antiphlogistin,Polygonum cuspidatum microemulsion gel and the control group had significant difference(P < 0.01)Results: 1.To study the preparation technology of Polygonum cuspidatum extract: Optimized of preparation technology of Polygonum cuspidatum extract preparation: 8 times the amount of 65% ethanol,90℃,extracting 2 times,each time 1h,extract 80℃ vacuum concentrated to the relative density of 1.20,80℃ by vacuum drying,process validation showed that extraction,concentration and drying process of Polygonum cuspidatum is stable and feasible.2.To study the prescription and process of Polygonum cuspidatum microemulsion gel: According to the single factor test and Box-behnken test,are as follows: Tween-80 microemulsion prescription of Polygonum cuspidatum,19.3% 1,2propylene glycol 7.7% and three glycerol acetate was 7.44%,borneol was 0.56%,distilled water was 65% and the content of Polygonum cuspidatum extract 1.4%;Polygonum cuspidatum microemulsion gel prescription: carbomer 940 1%,glycerol 10%,triethanolamine 1.4%,the 87.6% microemulsion of Polygonum cuspidatum.3.Evaluation of in vitro absorption of Polygonum cuspidatum microemulsion and Polygonum cuspidatum microemulsion gel:Microemulsion and microemulsion gel in vitro of Polygonum cuspidatum transdermal absorption evaluation by transdermal test results,and Polygonum cuspidatum microemulsion gel antiphlogistin compared Polygonum Q24 significantly increased,Qs significantly reduced,and compared with Q24 and Qs of Polygonum cuspidatum antiphlogistin significantly effective components of polydatin in Polygonum cuspidatum microemulsion decreased effective ingredient emodin the Q24 was not statistically significant.4.Local pharmacokinetics study of Polygonum cuspidatum microemulsion gel: Polygonum cuspidatum antiphlogistic cream and polydatin microemulsion gel polydatin.Emodin is a small t1/2 alpha in the anti-inflammatory cream of Polygonum cuspidatum.The distribution of drugs in mice is eliminated rapidly,and it is not easy to accumulate poisoning.5.Preliminary pharmacodynamic study of Polygonum cuspidatum microemulsion gel: In the experiment of anti-inflammatory,anti-inflammatory ointment,microemulsion gel of Polygonum cuspidatum and model group with significant difference(P < 0.01),analgesic experiments of Polygonum cuspidatum antiphlogistin,Polygonum cuspidatum microemulsion gel and the control group had significant difference(P < 0.01)Conclusion: The preparation process of Polygonum cuspidatum microemulsion gel is reasonable and feasible.The original preparation of giant knotweed anti-inflammatory cream and Polygonum cuspidatum microemulsion gel have certain anti-inflammatory and analgesic effects.
