The ultimate method for treating terminal organ disorder is organ transplantation.Human leukocyte antigens(HLAs)on the cell surface of donor organ are the major cause for organ transplant rejections.Although human embryonic stem cells(hESCs)have the ability to give rise to any type of somatic cell and are capable of unlimited proliferation,there is the risk of causing immune rejection.Therefore,immune-mediated rejection of hESCs greatly hinders its clinic application in organ transplantation.In this research,a human embryonic stem cell line with knockout HLA-A and HLA-B5101 was established via Transcription activator-like effector nuclease(TALEN),which is a new gene editing technique.Two pairs of TALEN was designed,which can specificly knockout HLA-A and HLA-B5101.Moreover,this study demonstrated HLA-A is knockout both at DNA level and protein level,and at DNA level,HLA-B5101 was also been knockout.The study will help promote the clinical application of human embryonic stem cells and the development of applications based on transplantation of human embryonic stem cells.
