Folic acid,as a kind of important B-group vitamins,possesses special guaranteeing effect for keeping organism normal physiologic functions.However,qualities of folic acid products in market are still being uneven and synthetic products can be harmful to people,which bring problems to both customers and regulators,just because of the absence of uniform detection method and quality standard.As an important kind of chemical raw materials,bisphenol A is widely used in China.However,the quality of products should be improved.Besides,trace amounts of bisphenol A in related products are threatening people's health,so it is essential to build up uniform detection method of bisphenol A.In this paper purification and purity determination of folic acid and bisphenol A were studied.Research on extraction of folic acid from powders of spinach leaves were carried out.Multiple parameters were studied based on extraction effects by single factor experiment coupled with orthogonal test,the optimized conditions were: solid-liquid ratio was 1:12,extraction solution was 30 mmol/L ammonium acetate,extraction temperature was 75 oC,extraction time was 30 min,absorbent in column chromatography was silica gel,eluent phase was dichloromethane–methanol(7:3),height of silica gel was 10 cm,loading capacity was 5 mL.The final extraction rate was 9.72 ?g/g.Qualitative analysis was made on the standard materials of folic acid prepared in laboratory through LC-MS,IR and NMR.Mass balance method was taken as basic quantitative method to determine the purity.The result was 90.64%,which was close to 90.57% determined by NMR method.And the overall uncertainty was 0.13%.Good stability for folic acid could be kept under 4 degrees in one month.Prep-HPLC,which was short for preparative high performance liquid chromatography,was applied in order to improve the purity of bisphenol A,the optimized conditions were: concentration of raw material was 20 mg/mL,mobile phase was 65:35 water and acetonitrile,elution time was 40 min,detection wavelength was 236 nm,flow rate was 15 mL/min,injection volume was 2.0 mL.The purity of product could be 99.92% and the yield was 21%,which could be provided as candidate reference material.Afterwards qualitative analysis was carried out through LC-MS,IR and NMR.The mass balance method was taken as basic quantitative method to determine the purity of bisphenol A material.The result was 98.63% and overall uncertainty was 0.45%.Good stability for bisphenol A could be kept under 4 degrees in one month.
