objective:Rheumatoid arthritis(RA),with joint pain and progressive failure as the main Clinical symptoms,is a long-term,chronic,systemic autoimmune disease.This disease is more common in females and its mainly age is concentrated in the 30-50 years old.And the average level of the prevalence rate in China is 0.2%-4%.The targeted therapy about the RA FSL transformation and proliferation,invasion,angiogenesis,proliferation of synovial pannus formation,cartilage destruction,bone erosion and degradation are also the hotspot studies.Biqi extract is widely applied in the treatment of RA,but its research on the mechanism of RA is not complete.So we act CIA rats as the animal model and observe the serum levels of IL-18,OPN,TNFalpha,COMP and OPN and the expressions of cartilage and synovium of OPN and COMP in CIA rats.To study the pharmacological effects of Biqi extract on CIA rats and look for new experimental evidence about the clinical treatment of RA.Methods:[1].CIA rat model was established and the swelling rate of the foot was observed and calculated.[2].Detecting the serum levels of TNF-? before drug administration by ELISA.Detecting the serum levels of IL-18,TNF-?,OPN,COMP after drug administration of 4W by ELISA.[3].Detecting the expressions of OPN and COMP in rats' synovium by IHC.[4].Detecting the expressions of OPN and COMP in the cartilage of rats by RT-PCR.[5].Pathological changes of synovium and cartilage were observed by HE staining.Results:[1].CIA rat model was established successfully.To observe the rat foot swelling rate,we founded the rat foot swelling rate in the model group increased significantly than in the normal group.Compared with the model group,the rat foot swelling rate decreased significantly than in the treatment group(P<0.05).[2].Comparison of cytokine serum levels: Compared with the normal group,the expressions of TNF-?,IL-18,OPN,COMP in the serum of the model group were higher than those of the normal group.Compared with the model group,the expressions of TNF-?,IL-18,OPN,COMP in the serum of the treatment group were lower than those of the model group.[3].Protein expression of cytokines in synovial tissue: The normal group synovial areas show a low expression of OPN.The normal group synovial no expression of COMP.The model group synovial areas show a large number of positive cells.The treatment group synovial areas show part of positive cells.Compared with the normal group,the expressions of OPN,COMP in the synovial areas of the model group were higher than those of the normal group.Compared with the model group,the expressions of OPN,COMP in the synovial areas of the treatment groups were lower than those of the model group.[4].Protein expression of cytokines in cartilage: Compared with the normal group,the expressions of OPN m RNA,COMP m RNA from rat cartilages in the model group higher than in the normal group.Compared with the model group,the expressions of OPN m RNA,COMP m RNA from rat cartilages in the treatment groups lower than the model group.[5].HE staining pathological observation: The pathological changes of synovial tissue and cartilage tissue were observed under microscope and HE staining.By using synovial tissue pathological score and the cartilage pathological score to calculate synovial inflammation.Compared with the normal group,the synovitis inflammation score and the cartilage pathological score in the model group were higher than in the normal group.Compared with the model group,the synovitis inflammation score and the cartilage pathological score in the treatment groups were lower than the model group.The middle dose group in inflammatory cell infiltration and synovial cell proliferation were decreased than the other treatment groups and the middle dose group in the injury degree of cartilage were better than the other treatment groups.Conclusion:[1].Biqi extract interventing the expressions of TNF-? and IL-18 in serum and blocking the formation of inflammatory cytokines network,thus inhibiting the development of arthritis.[2].Biqi extract the reducing the expressions of OPN in serum and the expressions of OPN and COMP in synovium and interventing synovial hyperplasia,thus inhibiting the formation of synovial angiogenesis and pannus.[3].Biqi extract the reducing the expressions of OPN and COMP in cartilage and interventing the degradation of cartilage matrix and cartilage destruction.[4].Compared with the model group,the serum levels of COMP in Biqi extract treated CIA rats lower than the model group,it reflects the treatment of CIA rats was effective.
