Background:Lung cancer is the commonest cancer and boasts of the highest mortality rate in the world.Chinese lung cancer patients account for more than 1/3 of them and rank first in morbidity and mortality in the world.The current treatments for lung cancer including operation,chemical therapy and radiotherapy bring obvious improvement to the survival of lung cancer patients,but the implementation and benefits of these treatments are undermined greatly by its drug resistance and high toxicity,so a safer and more effective method is urgently needed to extend patients' life.In the past 20 years,bio-immunotherapy has been considered a new “green therapy” and drawing more and more attention.Its principle of treatment is to amplify effector cells(such as NK cells,DC cells and CIK cells)in vitro,stimulate organism's immune response and depend on the high killing activity of effector cells and the immune system to fight against tumor cells.Objective:The clinical significance of Pro-GRP and T lymphocyte subpopulation of lung cancer patients receiving biotherapy for their immune functions is discussed by examining Pro-GRP,Tlymphocyte subpopulation and regulatory T cell Treg.Method:45 patients diagnosed with lung cancer by Tianjin Fifth Central Hospital from September 2014 to August 2015.And 40 healthy persons receiving physical examination are set as a healthy control group.CD3~+,CD4~+,CD8~+,Treg,CEA,NSE,CYFRA-211 and Pro-GRP in peripheral blood are examined.After three cycles of reinfusion of DC-CIK autologous cells,the same methods are adopted to collect the venous blood of patients to examine Pro-GRP,T lymphocyte subpopulation and Treg cells,the static analysis is conducted with SPSS19.0 software,multiple groups ofindependent samples are tested with single-factor variance,t test of the comparison before and after biotherapy is conducted with paired samples,correlation analysis is tested with Pearson and the job characteristics curve(ROC curve)of subjects is drew.P<0.05 is statistically significant.Results:Results:1.Pro-GRP,NSE,CEA and CYFAR21-1 have diagnostic values for the different pathological patterns of lung cancer,Pro-GRP and NSE have higher diagnostic values for SCLC,and ROC curve analysis thinks that the area below Pro-GRP curve has a higher Pro-GRP sensitivity than NSE,a higher specificity than NSE and higher diagnostic values for small cell lung cancer.2.The measured values of CD3~+,CD4~+,CD8~+ and CD4~+/CD8~+ of lymphocyte subpopulation in the peripheral blood of lung cancer patients are lower than that of control group.P<0.05 means the difference is statistically significant.The numbers of Treg cells in those groups are all higher than that of control group,which provides a scientific reference basis for the evaluation of cellular immune function of patients.3.After biotherapy,lung cancer patients' T lymphocyte subpopulations increase to different degrees,Treg cells decrease compared with before treatment,Pro-GRP in small cell lung cancer obviously decreases and non-small cell lung cancer group doesn't have obvious changes.4.Pro-GRP is negatively related to T lymphocyte subpopulation and has a significant positive correlation with Treg cells in different groups.P<0.05 means the difference is statistically significant,namely the measured values of T lymphocyte subpopulation and Treg cells increase with that of Pro-GRP.And it is indicated that when Pro-GRP increases,T lymphocyte lowers patients' immune function.Conclusion:1.Serum Pro-GRP can clinically work as the marker of lung cancer and provide reliable clinical basis for the early diagnosis of lung cancer.2.The expression level of T lymphocyte to some extent reflects lung cancer's ability to invade,spread and evade immunological surveillance and indicates a low immune function and lung cancer patients' immunosuppression.3.DC-CIK biotherapy can improve the clinical effects of ling cancer and enhance the immunocompetence of organism...
