Artemisia argyi is a traditional medicinal material used in China,it has a variety of pharmacological effects including antioxidation,antitumor,antibacterial and so on.It has numerous and complicated chemical substances and the main chemical components of volatile oil including flavonoids,tannins,eudesmane,polysaccharides,three terpenoids and trace elements etc..Along with the extensive research of Artemisia argyi and the growing maturity of the extraction technology,the chemical composition of the biochemical activity has been further studied.This paper aimed to study on chemical constituents of three extracts of Artemisia argyi,further study went on the basis of chemical constituents,to certified the anti-inflammatory effects of three extracts in vitro(inflammation of RAW264.7 macrophage model)and in vivo(zebrafish infection trauma inflammation model).First of all,the main peaks were identified by gas chromatography-mass spectrometry instrument,and the main chemical components were identified by referring to relevant literatures and CAS numbers.Via MTT,Griess,Real-time PCR experimental method to evaluated anti-inflammatory effects based on the chemical composition of three extracts both in vitro and in vivo.In vitro,the results showed out that 500ng/mL LPS stimulated RAW264.7 cells for 6h significantly increased the expression of inflammatory cytokines IL-6,IL-1?and TNF-? that proved the inflammatory models were successfully constructed;MTT method tested that the three extracts on cell toxicity are small,and showed low concentration on RAW264.7 cells proliferation.Three extracts showed IC50 were higher than 50?g/ml;Griess detection showed that the three extracts could significantly decrease the release amount of NO after macrophages stimulated by LPS,and showed a concentration-dependent decrease;Real-time PCR detection method found that three extracts can effectively reduce the expression and secretion of inflammatory factors IL-6 ? IL-1?and TNF-?after RAW264.7 stimulated by LPS.In vivo experiments,we observed that there were a decrease in the number of fluorescently labeled macrophages and neutrophils in the zebrafish model under fluorescence microscope.The three extracts of Artemisia argyi can be used as an effective drug for the treatment of inflammatory diseases because of its low toxicity and high efficiency.
