Objective Discussing the effects of resveratrol(Res)on the matrix metalloproteinase-2(mmp-2)activity and resistance to demineralization,in order to provide experimental evidence for the clinical application of resveratrol to protect demineralized dentin matrix.Methods Using a body fluid MMP-2 activity colorimetric assay kit to measure the effective concentration of resveratrol on the inhibition of odontogenic mmp-2.The surface morphology of dentin was examined by laser confocal microscopy and scanning electron microscopy.The degradation of dentin collagen fibers in each treated group was analyzed by enzyme-linked immunosorbent assay(ELISA)with human type I collagen type pyridine crosslinked peptide(ICTP).Results Resveratrol effect on the activity of matrix metalloproteinase-2 showed a certain relationship in a certain range of concentration,the concentration in 12 5-100umol/L,with the increase in drug concentration,the inhibition of MMP-2 increased significantly,but in 100-400umol/L,the inhibitory effect is relatively stable.The CLSM 3D images showed the demineralization depth of dentin slices: 100umol/LRes in the experimental group was smaller than that in the negative control group,and there was no significant difference between the positive control group(0.2%CHX).SEM was used to observe the morphology of dentin: The demineralization of dentin surface of 100umol/LRes treatment group was weaker than that of negative control group,which showed that the peritubular dentin demineralization was not obvious,and the opening degree of dentinal tubules was smaller than that in negative control group.And 100 umol / L Res group and 0.2% chlorhexidine group can be seen at different levels of granular sediment in the dentinal tubules.The results of ICTP kit,ie,the release of dentin ICTP showed that there was no significant difference between the Res group and the chlorhexidine group,and there was significant difference between the Res group and the chlorhexidine group after the simulated artificial caries formation process,ie,pH cycling.The release of ICTP in 0.2% chlorhexidine group was significantly lower than that in negative control group,which indicated that the amount of collagen fibers degradation in the negative control group was significantly higher than that in the Res group and the chlorhexidine group.Conclusions Resveratrol can inhibit the activity of matrix metalloproteinase-2,thereby inhibiting collagen degradation and dentin demineralization,play a role in protecting the demineralized dentin matrix...
