| Objective The chemical constituents of volatile oil and alkaloids of Zuojin Pill and Xianglian Pill were analyzed qualitatively by chromatographic-mass spectrometry.And then the constituents of Zuojin Pill and Xianglian Pill were studied by network pharmacology.At last to explore the mechanism of component-target-pathway for anti-diarrhea,anti-gastric ulcer and ulcerative colitis.Methods In this study,the volatile oil and alkaloids of Zuojin Pill,Xianglian Pill and constitutent herbs were separated and analysed by Gas chromatography-mass spectrometry and Liquid chromatography-mass spectrometry.The chemical constituents were identified by searching database and literature.Afterwards,to establish the multi-component-multi-target network by PharmMapper service system,TTD and DrugBank database,then the target was analyzed by GO enrichment and KEGG pathway annotation using MAS 3.0 software.The component-target-pathway network of Zuojin Pill and Xianglian Pill for anti-diarrhea,gastric ulcer and ulcerative colitis was constructed by Cytoscape software and analyzed.Results Volatile oil Ingredients: A total of 63 compounds were identified from Zuojin Pill,and 76 compounds were identified from Xianglian Pill.80,78,76 compounds were obtained from Coptis,Evodia and Radix.Alcoholic Extracts: Berberine,coptisine,jatrorrhizine,palmatine,evodiamine,rutaecarpine,xylenolide,and dehydroalinolactone were obtained from Zuojin Pill and Xianglian Pill.It was predicted that there were 30 active ingredients from Zuojin Pill acting on 10 targets,which were related to 27 pathways.There were 45 active ingredients from Xianglian Pill which were involved in 28 pathways acting on 12 targets.Conclusions In the study,the chemical constituents of Zuojin Pills and Xianglian Pills were comprehensively explained from two aspects of volatile oil and alcohol extract.The network model of Zuojin Pills and Xianglian Pills for anti-diarrhea,anti-gastric ulcer and ulcerative colitis showed the mechanism of multi-component,multi-target and multi-pathway.To provide a scientific basis for the further study of the material basis and pharmacological mechanism of Zuojin Pills and Xianglian Pills. |
