Objective: To investigate the expression of TWSIT1 and HIF-1? in human gliomas tissues and its correlation with survival prognosis,so as to provide a new method for the diagnosis and treatment of malignant gliomas Methods:56 cases gliomas collected in Affiliacted Union Hospital of Fujian Medical University from June 2010 to June 2014,10 cases of normal brain tissues.The expression of TWSIT1 and HIF-1? were detected by immunohistochemistry in 56 cases of gliomas and 10 cases of normal brain tissues.Through phone follow up patient's survival conditons.Using spss19.0 statistical methods for data analysis,Comparison of TWSIT1,differences in expression of HIF-1? in normal brain tissue and gliomas tissue analysis of TWSIT1,HIF-1?,the correlation between the pathological level of the three,and the clinical factors for survival analysis.Results: 1.TWSIT1 in normal brain tissue and low grade gliomas(I,II)was negative or weakly positive expression,while in high grade gliomas(III,IV)is mainly positive or strong positive expression.TWSIT1 has significant difference expression in normal brain tissue and gliomas tissue(p<0.001);the expression level was positively correlated with pathological grade(p<0.001).2.HIF-1? expressed in the nucleus and cytoplasm of gliomas and normal brain tissues.No expression of HIF-1? in normal brain tissues,and the low grade gliomas(I,II)was mainly negative or weakly positive expression,the high grade gliomas(III,IV)are mainly positive or strong positive expression.HIF-1? has significant difference expression in normal brain tissue and gliomas tissue(p<0.001);the expression level was positively correlated with pathological grade(r=0.553,p<0.001).3.TWSIT1 was positively correlated with the expression of HIF-1?(r=0.841,p<0.001)4.The single factor analysis showed that TWSIT1,HIF-1?,KPS,pathological grade,age,impact on overall survival;the multivariate analysis showed that TWSIT1,HIF-1?,pathological grade significantly affect the prognosis of gliomas.Conclusion: 1.The expression of HIF-1? is positively correlated with the expression of TWSIT1,and the expression of HIF-1? in gliomas tissue may promote the epithelial mesenchymal transition by regulating the expression of TWSIT1.2.TWSIT1,HIF-1?,and pathological grade were independent prognostic factors of gliomas patients.
