Objective:To investigate the effect of pirfenidone on proliferation,invasion and apoptosis of rectal cancer cells HCT15 and LOVO,and to explore the anti-tumor effect in rectal cancer cells.Methods:1.The effect of PFD on the proliferation of rectal cancer cells,HCT15 and LOVO was examined by CCK-8.The growth curve was drawed and the IC50 was calculated.2.The effect of PFD on the invasion of rectal cancer cells HCT15 and LOVO was examined by Transwell.The differences of the number of cells passing through the matrigel were found by taking pictures under the microscope and calculating directly.3.The effect of PFD on the apoptosis of rectal cancer cells HCT15 and LOVO was examined by Flow Cytometry.The relationship between PDF and apoptosis of rectal cancer cells was investigated by calculating the apoptotic rate.4.The effect of PFD on the proliferation,invasion and apoptosis of rectal cancer cells was detected by CCK-8,Transwell and Flow Cytometry again,respectively,after X-ray irradiation of rectal cancer cells HCT15 and LOVO.Results:1.The results of CCK-8: Rectal cancer cells or rectal cancer cells undergoing X-ray irradiation showed slow growth and inhibition of proliferation at various time points(24h,48 h,72h)after PFD treatment,and with the prolongation of drug action time or concentration increased,the cell growth inhibition rate also increased.2.The results of Transwell: The number of rectal cancer cells or X-ray irradiated rectal cancer cells passing through the matrigel were significantly decreased after PFD treatment,and there were statistical differences(P <0.01).3.The results of Flow Cytometry: The number of apoptotic rectal cancer cells or X-ray irradiated rectal cancer cells were significantly increased after PFD treatment,and there were statistical differences(P <0.01).Conclusion:1.Pirfenidone can inhibit the proliferation of the rectal cancer cells HCT15 and LOVO and cells after X-ray irradiation.2.Pirfenidone can inhibit the invasion of the rectal cancer cells HCT15 and LOVO and cells after X-ray irradiation.3.Pirfenidone can promote the apoptosis of the rectal cancer cells HCT15 and LOVO and cells after X-ray irradiation.
