Objective To find out the change of gene expression and explore the mechanism of slicing factor SRSF3 in the apoptosis of hepatocellular carcinoma line induced by TPL.Method Grouping:normal hepatocytes line L02 group and hepatocellular carcinoma cell line Hep G2 untreated group,treatment group was Hep G2 treated by TPL with different concentration.Western blot to detect the change of expression of the SRSF3 protein.The apoptosis rate of cell in treated group was detected by flow cytometry through Annexin V/PI double staining.The change of expression of the SRSF3 gene was detected by RT-PCR.Result:Human hepatocellular cell Hep G2 have been inhibited in proliferation when treated by TPL,and the inhibitory effect has dose effect relationship.The half maximal inhibitory concentration(IC50)of TPL acted on Hep G2 cell for 72 hours is 3..85±1.24ng/ml.Flow cytometry results indicate that TPL induce apoptosis in Hep G2 cell and it has dose-effect relation within certain range.Western blot results show the expression of SRSF3 protein in human normal liver cells is lower than in human hepatocellular cell Hep G2.When treated by different concentration of TPL for 72 hours,the expression of SRSF3 protein shows a decline trend with the increased concentration in Hep G2 cells.The results of fluorescence quantitative PCR indicate that:Compared with human normal liver cells L02,the SRSF3 m RNA expression relative value is signifiacantly higher in Hep G2 cell group;when treated by different concentration of TPL for 72 hours,the SRSF3 m RNA expression of treatment group shows to descend.Conclusion: TPL inhibit Human hepatocellular cell HepG2 cell proliferation and induce its apoptosis.SRSF3’s high expression may be involved in the TPL-induced apoptosis.
