Objective:To investigate the effects of high fatty acid and high carbon acid on the calcification and growth of xenograft model of HCC827 human lung adenocarcinoma.Methods:Cultivate HCC827 Human non-small cell lung adenocarcinoma cells to a certain number,ensure that cells do not contaminated,logarithmic phase,vitality and growth state good cells in serum free medium heavy suspension and counting cell concentration 1 * 107 / ml,then subcutaneous injection of cancer cells,0.2ml HCC827 single cell suspension vaccination in skin of the mice,the weeks of nude mice about 4 weeks,and regular observation and measurement of tumor growth.For tumor tissue to about 10-15 mm in diameter,select forty xenograft model of HCC827 human lung adenocarcinoma of nude mice,and randomly divided into 4 groups,each group hanve 10 nude mice,group of oleic acid intratumor injection,group of sodium bicarbonate intratumor injection,group of oleic acid and sodium bicarbonate intratumor injection and group of normal saline intratumor injection.Control group tumor tissues injection of oleic acid 0.2 ml,respectively,NaHCO3(concentration for 595 mmol / L)0.2 ml,oleic acid 0.1 ml + NaHCO3(concentration for 595 mmol / L)0.1 ml and 0.2 ml normal saline,Observing the changes of tumor volume and the case of calcium deposition.Results:At the end of the experiment,the volume of transplanted tumors of four groups in order of size-small to large is group of oleic acid,group of oleic acid + sodium bicarbonate,group of sodium bicarbonate and group of normal saline.Group of Oleic acid,in the process of the experiment,there are seven transplantation tumor ulcerate through the skin,group of oleic acid + sodium bicarbonate has four transplantation tumor ulcerate through the skin,and group of sodium bicarbonate and group of normal saline were have three transplantation tumor ulcerate through the skin;And that,in the process of the experiment,the volume of the group of oleic acid is most unstable,the volume has suddenly growth trend,also has a tendency to suddenly decrease.Alkaline phosphatase(ALP)staining showed four groups are alkaline phosphatase staining positive,but the staining intensity,from strong to weak was group of oleic acid,group of oleic acid + sodium bicarbonate,group of sodium bicarbonate and group of normal saline.HE staining showed that group of oleic acid and group of oleic acid + NaHCO3 there was an obvious calcium salt deposition,group of Na HCO3 and group of normal saline did not see obvious calcium salt deposits.And the transplanted tumor tissue pathological slices of group of oleic acid and group of oleic acid + sodium bicarbonate showed inflammatory necrosis and purulent necrosis,the transplanted tumor tissue of group of sodium bicarbonate and the group of normal saline no necrosis of tumor tissue.Calcium salt dyeing showed that group of oleic acid and group of oleic acid + NaHCO3 there was an obvious calcium salt deposition,group of NaHCO3 and group of normal saline did not see obvious calcium salt deposits.Conclusion:Local high oleic acid can promote tumor tissue calcium salt deposition,and can restrain the growth of the tumor.The volume of the group of oleic acid has sudden growth trend and sudden decrease trend,all of also suggests that local high oleic acid easily make tumor tissue necrosis,oleic acid can inhibit the growth of tumors with oleic acid also reduce tumor tissue necrosis in tumor tissues.Separate local high sodium bicarbonate may no obvious role in promoting tumor calcification,but local high sodium bicarbonate inhibits tumor growth,and obvious effect,but the tumor volume will continue to grow after stop using sodium bicarbonate,may be associated with sodium bicarbonate changed tumor partial acid environment,make the local microenvironment be bad for tumor growth,after stop using NaHCO3,local microenvironment of tumor tissue recovery for acidic microenvironment,which in turn promote the growth of tumor tissue.
