Objection:There were rich resources of Rose xanthina Lindl.in Shanxi,but the utilization rate of Rose xanthina Lindl.fruit was not very well.In this thesis,we studied the antioxidant and anti-thrombotic effect of Rose xanthina Lindl.fruit extraction,it provided rich experimental basis for the next development of the use of the Rose xanthina Lindl.fruit as a healthy food.Methods:The antioxidant activity was measured with the methods of DPPH· scavenging,pyrogallol autoxidation and phenanthroline by using ascorbic acid as the positive control.Total flavonoids was measured with the methods of Sodium nitrite-aluminium nitrate chromogenic,and phenolic compounds was determined by Foline-reagent method.The D-galactic model was used in mice to explore antioxidant effect in vivo.MDA and SOD in serum and liver homogenate of mice was determined to evaluate the antioxidant effect of Rose xanthina Lindl.fruit.The mice were randomly allocated into five groups,including control group,the low-dose group,medium-dose group,high-dose group,the positive control group.The tail-thrombus models were established by carrageenan after the mice were fed for 5 d.The length of dark tails was observed after the models were established for 24 h and 48 h.The mice randomly divided into 5 groups as aforementioned.All groups were treated with drug or NS.At the dosage of 0.2 mL/10 g once daily for 7 consecutive days to observe the effect of Rose xanthina Lindl.fruit extraction on the formation of pulmonary thromboembolism.The ADP-induced platelet aggregation was measured by using multifunctional platelet aggregometer after the different concentration of the alcohol extract of Rosa xanthina Lindl.fruit was added into rat platelet rich plasma respectively.The prothrombin time(PT),thrombin time(TT)and activated partial thermoplastic time(APTT)of human plasma dried were determined at different dose groups of extract of Rosa xanthina Lindl.fruit in vitro.At 0,4,8,24 h,the blood clots quality were weighed at different dose of extract of Rosa xanthina Lindl.fruit,and the blood clots dissolution were calculated.Results:The results showed that the extracts had certain antioxidant activity,and exhibited a good concentration-response relationship.Different polarities had different abilities on scavenging free radical,in which the ethyl acetate fraction of the fruit had the most potent free radical scavenging activity on DPPH·,O2-·,·OH,with IC50 18.52,737.00,605.50?g/mL,respectively.In antioxidant experiment in vivo,SOD activity of liver homogenate and serum SOD activity of high,medium and low dose groups compared with model group was significant increased(P<0.01).Compare with the control group,the relative black tail length and relative black tail length inhibition rate were significantly reduced in the high-dose group and medium-dose group(P<0.01).Compare with the control group,the high-dose group and medium-dose group significantly increased the survival rate of mice(P < 0.05 or P < 0.01)and the protective rate were 50.0% and 58.3% respectively.Compared with the control group,the platelet maximum aggregation was significantly reduced in the high-dose group,medium-dose group and low-dose group of Rose xanthina Lindl.fruit(P < 0.01).Compared with the control group,the extract of Rose xanthina Lindl.fruit was able to prolong APTT,TT,PT(P < 0.01).At the 4,8,24 h,the blood clots dissolution were significantly increased in extract of Rosa xanthina Lindl.fruit in high-dose and medium-dose groups.Conclusion:The ethyl acetate of the fruit is a potential natural antioxidant,It also can be further developed into an antioxidant functional food.The extract of Rosa xanthina Lindl.fruit demonstrates good anticoagulation and antithrombotic effects.
