Objective: To investigate the inhibitory effect of resveratrol on gastric cancer cells and inhibit the expression of DNA methyltransferase and histone deacetylase.Methods: Culture gastric cancer cells with difference conditions(blank control;25μmol/L,50μmol/L,100μmol/L DMEM include Res)for 24 hours,the cell apoptosis and cell cycle was assayed by flow cytometry,the expression of DNMT1、DNMT3a、DNMT3b、HDAC2、HDAC11 was assayed by immunocytochemisty.Results: Culture after 24 hours,all groups use flow cytometry,the test result of cell apoptosis respectively 1.48±0.15、12.77±0.26、26.59±0.43、38.59±0.23,F=9585.25,p<0.01;the test result of cell cycle respectively 44.47±0.41、52.15±0.34、55.22±0.32、71.48±0.40,F=2807.43,p<0.01;the expression of DNMTs and HDACs tested by immunocytochemisty respectively DNMT(191.70±0.60、 197.37±0.73、201.88±0.29、207.97±0.81,F=347.21,p<0.01)、DNMT3a(187.64±0.96、197.21±0.63、203.35±0.74、208.17±0.30,F=481.49,p<0.01)、DNMT3b(191.69±0.65、196.36±0.44、201.04±0.37、205.69±0.46,F= 452.17,p<0.01)、HDAC2(184.58±1.01、193.32±0.57、200.60±0.84、205.12±0.53,F= 414.68,p<0.01)、HDAC11(189.94±0.70、194.14±0.99、201.71±0.73、207.46±0.77,F= 445.69,p<0.01).Conclusion:1.Resveratrol inhibit the expression of DNA methyltransferase and histone deacetylase can promotes gastric cancer cell apoptosis,and to block gastric cancer cells in G1 phase.2.The effects of resveratrol anti-cancer with concentration dependent. |
