Objective:To establish animal models of oral ulcer and observe the effects of a non-thermal atmospheric pressure argon plasma jet on the healing time,and the levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in oral mucosal ulcer of golden hamsters,and discuss the feasibility of the local use of NTAPs in oral ulcer,provide reference basis for potential clinical application.Materials and methods:1.The animal models of oral ulcer were established by the use of oxygen free radicals,the shape and size of the ulcer were observed by eyes and The oral mucosa was harvested for HE staining for pathological observation when ulceration.2.56 animals were randomly divided into: the first batch modeling group(n=48),the second batch of modeling group(ulcer model group,n= 4,modeled 72 h after the modeling of the first batch animal model)and normal control group(n=4,normal,do not have any processing).The first batch modeling group is divided into: natural healing group,the drug group,2 argon groups(30 s,120 s)and plasma groups(30 s,120 s),8 of each group.The shape and size of the ulcers were recorded after the ulceration,and all of the treatments were taken after the ulceration,1 times a day,for 3 days.24 h after the end treatment,72 h after the second batch animal model were modeled,4 animals selected randomly from each group were sacrificed for the detection of S0 D activity and MDA content in the mucosa ulcer;the healing time of the ulcers the remaining 24 animals were observed and recorded,and then all of the remaining animals were sacrificed for the detection of S0 D activity and MDA content after the ulcers healed.Results:1.the mucosal ulceration was observed after 72 hours,round or class round,about 0.5-0.6 cm in diameter,covered by pale yellow pseudomembrane,the surrounding hyperemia,edema,the microscopic pathological characteristics were similar to human oral ulcer.2.(1)average area of the ulcers(0.216±0.014)cm2,there was no significant differences between each groups(P > 0.05);healing time of mucosal ulcer: the healing time of plasma groups was obviously shorter than the rest groups,the differences were significant(P = 0.00),but there was no significant difference between 30 and 120 s plasma groups(P > 0.05).(2)compared with normal control group,after the modeling of the ulcer model group,SOD activity decreased significantly(P = 0.00),MDA content increased significantly(P = 0.00).24 h after the last processing,there was no significant differences in the SOD activity and MDA content between plasma groups and normal control group(P > 0.05),but there was significant difference when compared with the ulcer model group and other groups(P = 0.00).There was no significant difference between the two plasma groups(P > 0.05).(3)There was no significant difference in SOD activity and MDA content between the normal control group and other groups after ulcer healing completely.Conclusions:1.The experimental results show that the animal models of oral ulcer established by the use of oxygen free radicals were success,and the ulcers are similar to human oral ulcers in clinical and pathology features.2.The experimental results show that:(1)there was no significant difference in ulcer size;(2)SOD activity in areas of mucosal tissue decreased significantly after the ulceration,MDA content increased obviously.24 h after the last treatment,30 s,120 s plasma treatment can increase S0 D activity and reduce MDA content effectively,to make them back to normal.And the healing time of 30 s,120 s plasma groups was obviously shorter than the rest groups.(3)With the method to establish animal model of oral ulcer,the SOD?MDA levels can return back to normal after ulcers healed completely.
