ObjectiveThe incidence of multiple myeloma(MM)ranked third in the blood tumors.The remission rate of disease treatment greatly improved with the application of new drugs,improvement of chemotherapy and maturation of stem cell transplantation,but the diseaserelapsedin a considerable number of patients in several months or years.Studies have shown that MRD-induced recrudescence is one of the reasons for its difficult to cure.Therefore,this medicine should be found,which can inhibit the migration and invasion of myeloma cells and be low toxicity,and plays an important role in therapy.This study was to investigate the effect of fucoidan on autophagy,migration and invasion of humanmultiple myeloma U266 cells,and to provide a theoretical basis for the clinical application of fucoidan in the treatment of multiple myeloma.Methods The U266 cells treated with fucoidan were cultured in vitro.The formationofautophagosomes was observed by transmission electron microscopy(TEM).Transwell assay was used to evaluate the effect of fucoidan on migratory and invasive abilities of U266.The protein levels of LC3-? /LC3-?,Beclin-1,P62,MMP9,CXCR4,p-AKT/T-AKT,p-mTOR/T-mTOR were detected by Western blotting.MMP9 concentration in the culture medium was examined by ELISA.Results Autophagosomes increased in fucoidan-treated cells compared with control group under TEM.Migratory and invasive abilities were inhibited by fucoidan in dose-dependent manner,which were suppressed by chloroquine(CQ).Western blotting demonstrated that expression ofLC3-? /LC3-?,Beclin-1 andMMP9 increased in fucoidan-treated cells,while P62,CXCR4,p-AKT/T-AKT and p-mTOR/T-mTOR decreased compared with control group.The result of ELISA showed that MMP9 concentration in the culture medium of fucoidan-treated cells significantly decreased.ConclusionFucoidan induces autophagy and inhibits migration and invasion in U266 cells.
