Side Effects Of Pramipexole In Parkinson’s Diesase:System Evaluation

Objective:To evaluate the safety of clinical application of pramipexole,especially the cerebrovascular disease risk,we used meta analysis method in the treatment of Parkinson ’s disease(PD)about side effects of pramipexole for evaluation system。Methods:According to the aim of research,after formulating the strict inclusion and exclusion criteria,we used computer to retrieve English database,such as: MEDLINE,PubMed,EMBASE,Google etc,and Chinese database,such as: CNKI,Wanfang,Cochrane Library etc,to find all randomized controlled trials for treatment of PD with pramipexole.The retrieval deadline is to February 2016.And then the meta analysis was performed by Review Manager 5.3 software on all the documents,which met the inclusion and the exclusion standard,and is in line with the modified Jadad score greater than or equal to 5 points.Result: This study Included 11 randomized controlled trials for the treatment of PD about pramipexole.We made a system evaluation for a high incidence of dizziness,headache,nausea in seven adverse reactions find that the pramipexole group were higher than the control group in all of the incidence of adverse reactions,.Among them,its analysis results showned the incidence of drowsiness,insomnia and different movement disorder of the pramipexole group risk is higher than that of the control group,with statistical significance,.And although the results of the analysis showed the headache,dizziness,orthostatic hypotension,nausea and other adverse reactions of pramipexole group risk is higher than the of the control group,the result was no statistical significance,.we considered these adverse reactions produce may have nothing to do with the role of the drug itself,being related to the choice of research object and the progression of the PD.In addition,it is important to note that all 11 articles in the literature,in addition to Ying,Wang mentioned serious adverse events such as the literature and cardiovascular events,the rest did not mentioned documents Such as fatal arrhythmia,acute coronary syndrome,acute cerebrovascular accident,brain heart syndrome and other serious complications and kidney disease of heart head blood-vessel complication.It showed the cardiac adverse reactions about pramipexole may be an individual case,which did not have universal significance.Conclusion: In the treatment of PD with pramipexole,the occurrence of adverse reactions,such as the drowsiness,insomnia,different movement disorder,appeared the increased risk,but the occurrence of adverse reactions,such as the headache,dizziness,orthostatic hypotension,nausea,appeared no obvious higher risk.Heart risk was only individually reported.Because this article existed obvious variety bias into the study of literature,and relatively high heterogeneity,and at the same time the literature number is relatively small,especially the size of the sample distribution,the analysis result needed more large sample randomized controlled trials to further verification,.And its conclusions were only for reference.
