Background: The morbidity of thyroid cancer had grown rapidly all over the world recent years,the papillary thyroid cancer accounted for more than half of all pathological types of thyroid cancer.Although the pathogenesis of thyroid papillary cancer is still not very clear,almost all of the malignant tumors are abnormal cell cycle regulation.Analysis of the DEPDC1 protein which regulate the cell cycle with thyroid papillary cancer and the mutated BRAF gene which is the most important molecular markers of thyroid papillary cancer is of great significance.Objective:To investigate the expression of DEPDC1 in thyroid papillary cancer and peritumoral thyroid tissues,to study the expression of DEPDC1 in the BRAF gene mutation type tumor tissues and the BRAF gene wild type tumor tissues,analysis the relationship between DEPDC1 with thyroid papillary carcinoma and the BRAF gene and clinical features.To discuss whether DEPDC1 can provide experimental basis for clinical diagnosis and therapeutic targets.Methods:Adopted on the basis of the hospital clinical specimens of contrast research,proposed use of the method of molecular epidemiology,existing specimen is utilized to extract protein and making the pathological section.Application of immunohistochemical method to detect the expression location of DEPDC1 in tumor tissue and adjacent normal tissues.Using Western blot method to detect the differences expression of DEPDC1 in the tumor tissues and adjacent normal tissues,in the BRAF gene mutation type and the BRAF gene wild type,in different clinical features groups.Results:Immunohistochemistry shows DEPDC1 expression in thyroid papillary cancer tissues and adjacent normal tissues.Detected in 5 cases of tumor tissue pathological sections,negative expression in 1 case,strong positive expression of 4 cases,the 2 cases of adjacent normal tissues were weakly positive expression.DEPDC1 was located in the nucleus and cytoplasm.Western blot results show DEPDC1 protein in thyroid papillary cancer tumor tissues is expressed for(59/62),Western blot results show DEPDC1 protein in peritumoral tissue is expressed for(14/20).Western blot results show DEPDC1 protein expression in thyroid papillary cancer tissues for(1.60±0.82),significantly higher than the DEPDC1 protein expression in normal tissue adjacent to tumors quantity(0.58±0.19)for the difference was statistically significant(P(27)0.05).DEPDC1 protein expression in the BRAF gene mutation type for(1.83±0.80),significantly higher than the BRAF gene wild type(1.03±0.45)for the difference was statistically significant(P(27)0.05).Male and female with DEPDC1 protein expression in tumor tissue volume,respectively(1.41±0.71),(1.64±0.83),the difference has no statistical significance(P>0.05).Patients with age greater than or equal to 45 years old and younger than age 45 DEPDC1 protein expression in tumor tissue volume,respectively(1.51±0.67),(1.64±0.91),the difference has no statistical significance(P>0.05).The experimental results show that DEPDC1 protein can be detected in thyroid papillary cancer tissue and normal tissue adjacent to tumors,and tumor tissue expression intensity was significantly higher than normal tissue adjacent to tumors.DEPDC1 protein expression level with thyroid papillary cancer of the BRAF gene mutation type specific molecular markers were positively correlated,and DEPDC1 protein expression and clinical characteristics such as gender,age,no significant correlation.Conclusion:DEPDC1 protein localization in the nucleus and cytoplasm of thyroid papillary cancer.DEPDC1 expression positive correlated with thyroid papillary cancer and BRAF gene mutation.DEPDC1 expression has no correlation with gender and age.
