Resveratrol Attenuates Neurologic Deficit And Neuroinflammation After Intracerebral Hemorrhage

Background: Natural antioxidant resveratrol(RESV)has been showed to improve histopathological and behavioral outcomes after various central nervous system diseases.However,there is no report on its neuroprotective effects on the secondary brain injury after intracerebral hemorrhage(ICH).Objective: The aim of this study is to evaluate the neuroprotective function of resveratrol after ICH.Methods: Male Sprague-Dawley rats were randomly divided into three groups: Sham group,ICH group and ICH+RESV group.Rats were intraperitoneally injected with drugs daily.Rotarod test and open field test were used to evaluate the recovery of motor disturbance pre-operation and post-operation.Rats were sacrificed after the last behavior test.Neuron injury and cell death in the hippocampal regions were measured by Nissl staining,and microglia activation in the cortex was detected by immunofluorescence staining of Iba-1.Images were captured on a fluorescence microscope coupled to a cooled charge-coupled device camera and processed by using the capture program for quantitative analysis.All the experimental data were analyzed using statistical software.Results: Compared with ICH group,rats that were treated with resveratrol for two weeks performed significantly better in behavior tests.In rotarod test,the duration on the rod was obviously decreased in ICH group and ICH+RESV group on the first day post-surgery,compared to their performance before the operation.The recovery was observed on the 7th day post-surgery in ICH+RESV group but 14 th day post-surgery in ICH group.Although both ICH group and ICH+RESV group showed significant recovery on 14 th day post-surgery,ICH+RESV group has striking performance compared to ICH group.Compared to sham group,the significant differences for distance traveled,average speed and rotations were observed on the first day post-surgery in ICH group and ICH+RESV group.After one week administration of resveratrol,the total distance and average speed in ICH+RESV group were increased significantly compared to ICH rats.Itindicated a partial recovery effects on motor ability of rats under resveratrol treatment.There were less neural damages in the hippocampus and decrease of microglia activation in ICH+RESV group.The number of neurons was obviously decreased after ICH surgery.After the treatment of resveratrol,the damaged neurons were reduced and the numbers of normal neurons were increased.The number of microglia cells was increased after ICH.However,the administration of resveratrol post to the operation led to a decreased number of Iba-1 which was up-regulated by ICH.Conclusion: Our results indicate that resveratrol is beneficial to alleviate the secondary brain injuries after ICH.
